Dendrobium is a rare Chinese herbal medicine.It has been proved by modern pharmacology that it possesses various biological activities such as antioxidation,anti-tumor,immune regulation and neuroprotection.Its main components include alkaloids,flavonoids,terpenoids,and polysaccharides,Amino acids,etc.Alkaloids are substances with potential antitumor activities,and are great promising with high efficiency and low toxicity.But,little report about their role in anticancer activity in Dendrobium was currently found.Therefore,it is necessary and urgent to develop alkaloid anti-tumor compounds from Dendrobium.China is a region with high incidence of gastric cancer,and great significance will have to study the inhibitory effect of Dendrobium on the proliferation of gastric cancer cells.In order to investigate the inhibitory effect of Dendrobium on gastric cancer cells and its potential alkaloid antitumor compounds,three types of Dendrobium(Dendrobium officinole,Dendrobium moniliforme and Dendrobium huoshanense)were selected as raw materials in this study.The results are a as follows:(1)The inhibition effect of each component on gastric cancer cells was detected by MTT assay.The results showed that among which the inhibition of D.officinale and D.moniliforme was the strongest with the IC50 value is 0.160 mg/m L and 0.149 mg/m L,respectively.(2)Annexin V-FITC/PI double staining method showed that raffinate phase from Dendrobium officinale can inducing apoptosis of SGC-7901 cells;q RT-PCR results showed that raffinate phase can down-regulated expression of m RNA of bcl-xl,up-regulated expression of m RNA of bak,bax,caspase-3,and caspase-7.Both speculate that the raffinate phase induces apoptosis by activating the caspase signal transduction pathway.(3)LC-MS detection found that D.officinale contains 36 alkaloids,of which sanguinarine has been reported to have anti-tumor activity.(4)Qualitative and quantitative analysis of sanguinarine of D.officinale in raffinate using HPLC method showed that the D.officinale in raffinate contained sanguinarine at a content of 150 ng/mg.MTT assay was performed to test the effect of sanguinarine on the growth of gastric cancer cell line SGC-7901,with IC50 values were 2.7 and 1.7μmol/L following exposure to sanguinarine for 24 and 48 h,respectively,suggesting that sanguinarine had a good inhibitory effect on SGC-7901 cells.The above results indicated that extracts from D.officinale and D.moniliforme have a stronger inhibitory effect on gastric cancer cells,and alkaloids are rich in D.officinale,which can provide reference for the screening,identification and development of alkaloids in D.officinale anti-tumor compounds.
