Objective:To investigate the change of the diversity of intestinal flora and the metabolic function analysis of the rats in the physiological state、the euthyroxhe heat syndrome and cold syndrome in cold water to decide the Scutellaria Baicalensis’s properties.Methods:The use of three parallel experiments,the physiological state、the euthyroxhe of heat syndrome and cold syndrome in cold water,60 male SD rats,then the rats were randomly divided into 6 groups,10 rats in each group,the blank group(K),the physiological state of full component group(SmQ),heat syndrome group(RmM),heat syndrome of full component group(RmQ),cold syndrome model group(HmM)and cold syndrome of full component group(HmQ);As it is done in the same experimental state,the blank group is a common component that can be used to compare,then divided into three groups,the physiology state,the heat syndrome state,the cold syndrome state;The blank group was given normal saline lml/100g,euthyroxhe of heat syndrome lml/100g,and the ice water bath was made for stiff limbs;the full component of Scutellaria baicalensis was 1ml/100g,after 16d,the rats were anesthetized and their cecum quick-frozen for later analysis.The 16S rRNA high throughput gene sequencing technology was applied to detect the caecum in rats.Illumina MiSeq platform of the original double end sequencing data;eliminating doubt sequence and high quality sequence statistics;then analysis of intestinal flora diversity.The intestinal flora of OTU numbers、common OTU analysis,Rarefaction curve,Species accumulation curves,Rank abundance curve,the Alpha diversity in richness index and diversity index,the taxonomic component analysis of the flora,PCA analysis of Beta diversity in the community,PLS-DA analysis,significant difference taxonomy and potential metabolic function analysis were carried out.Result:Base on the Illumina Miseq platform,compare the physiological state blank group and full component group(SmQ);blank group,heat syndrome group(RmM),heat syndrome of full component group(RmQ);blank,cold syndrome model group(HmM)and cold syndrome of full component group(HmQ),find that the three state that the rarefaction curve,species accumulation curves,and rank abundance curve are adequately to reflect the richess and diversity;The community fuly reflect the diversity of the sequencing depth of current sample,the number of OTU close to the actual situation,throught the analysis of PCA of Beta diversity in the samples were found that community similarity difference was little,different samples appreared significant differences in community structure,indicating that significant impact to the community structure after model and give medicine.Through the comparative analysis of community and key species selection,in normal group,heat syndrome,cold syndrome group of different components in the sample in the community were significantly separated,in the same group in the sample community close to each other,the above can explain what we have samples adequately used to analyze the changes in community different syndromes of intestinal flora.In the physiological group,after intervention of Scutellaria Baicalensis,the number of OTU decrease;richness index and diversity index significantly decrease,compared with K and SmQ,there is a decreasing trend in the species of phylum and genus,the phylum level is primary composed of Bacteroidetes and Firmicutes,the amount of Bacteroidetes descend,the number of Firmicutes(Firmicutes)increased,but not obvious;the significant differences in genus are Blautia、Dorea、Helicobacter;for cell motility,membrane transport,translation,energy metabolism showed inhibitory trend,sugar metabolism in the metabolism show promote trend.The number of OTU in heat syndrome model group increased,and the number of OTU decreased after the treatment,RmM group richness index significant increased,after give dose,RmQ group richness index significant decreased,indicating that the flora of heat syndrome rats increased significantly,and the models were successful.However,after giving Scutellaria baicalensis,the abundance and diversity of the cecal intestinal flora in rats changed significantly;compared with RmM and K,the amount of phylum and genus increase,after intervention,the phylum and genus decrease;compared K、RmM and RmQ,The obvious phylum is Deferribacteres,the phylum level is primary composed of Bacteroidetes and Firmicutes,after modeling,the amount of Bacteroidetes increase,after give medicine decrease;the amount of Firmicutes decrease,after giving medicine increase but not obvious;the significant difference genus is Mucisirillum;cell motility,membrane transport,glucose metabolism and carbohydrate metabolism were promoted.Scutellaria baicalensis showed a trend of inhibition after intervention,but not obvious on energy metabolism.In the cold syndrome group of ice water,the number of OTU in the model group decreased and the number of OTU decreased after the drug.Compared the Alpha diversity index in the three groups of K,HmM and HmQ rats,indicating that richness index in HmM group was significantly lower than that of K group,drug intervention group HmQ richness index is further reduced;the diversity index of the HmM group was lower than K group,but not significantly,that cold syndrome rat model of bacteria community diversity is small,given baicalin after treatment,rats blind intestinal microflora community diversity did not change significantly.It is suggested that Scutellaria baicalensis can reduce the abundance of intestinal flora in cold syndrome model rats,but it does not change its diversity.Comparison of K,HmM,HmQ,significant difference phylum is Deferribacteres,phylum level is mainly composed of Bacteroidetes and Firmicutes,after modeling Bacteroidetes number decreased after administration decreased;after modeling Firmicutes(Firmicutes)increased,but not significantly increased after administration;The significant difference phylum is Christensenella、Biophila;The cell motility,membrane transport,glucose metabolism and carbohydrate metabolism were inhibited.The effect of Scutellaria baicalensis injection was more obvious after intervention,and the effect on energy metabolism was not obvious.Conclusion:The results of the above experimental results show that Scutellaria can significantly inhibit the number of OTU,richness index,diversity index and community structure of intestinal flora;It is found that Scutellaria can reduce the formation of sugar,promote the synthesis of glycogen,inhibit the production of short chain fatty acids,and promote the synthesis of fat through the analysis of the distribution of saliency and genera.Through the analysis of microbial metabolic function,the most important metabolic pathway of Scutellaria is the carbohydrate metabolism and carbon water in material metabolism.Compound metabolism,combined with previous experimental results in this project,Scutellaria baicalensis has inhibitory effects on metabolism and energy metabolism related metabolic pathways.and it can inhibit the material metabolism at the same time,From the angle of intestinal microflora,conclude that the Scutellaria Baicalensis’s drug property is cold.
