| PurposesBy studying the effects of goose blood and serum on human gastric cancer BGC-823 cells,the anti-tumor components of goose blood were revealed.In order to further clarify the anti-cancer effect of goose blood.Methods1.Preparation of whole blood and serum from goose blood.2.Cell culture:human gastric cancer BGC-823 cells,human esophageal cancer ECA cells.3.The effects of different concentrations of goose blood and serum on the proliferation of human gastric cancer and esophageal cancer cells were detected by CCK8 method.4.ELISA kit was used to detect the tumor markers such as Mmp7、mmp2、MT1-MMP、tnfα、il-6、il-8.5.The effects of different concentrations of goose blood and serum on apoptosis of human gastric cancer BGC-823 cells were detected by flow cytometry.6.The expression of Caspase-3mRNA in human gastric cancer BGC-823 cells was detected by RT-PCR technique.Research Result1.CCK-8 method shows that the whole blood and serum of goose blood in different concentrations can obviously inhibit the proliferation of human gastric cancer BGC823 cells,but it has no significant effect on the ECA cells of human esophageal cancer.2.The results of ELISA kit showed that the difference of whole blood and serum in different concentrations of goose blood was significant compared with the control group(P<0.05).The antitumor effect of the whole blood and serum of different concentrations of goose blood was slightly inferior to that of the positive drugs,but they all had obvious antitumor effects.3.The results of flow cytometry showed that different concentrations of goose blood and serum could significantly induce apoptosis of human gastric cancer BGC823 cells compared with control group.And when the concentration is 15%,the effect is remarkable.4.The results of RT-PCR detection showed that:compared with the blank group,the positive drug group,the serum group and the whole blood group could significantly up-regulate the expression of Caspase-3mRNA,and the difference was significant(P<0.05),and compared with the positive group,the expression of Caspase-3mRNA was decreased in the goose blood serum group and the goose blood whole blood group.There was significant difference(P<0.05)and there was no significant difference between goose blood group and goose blood serum group,and there was no significant difference between goose blood whole blood group and goose blood serum group(P<0.05),and there was no significant difference between goose blood whole blood group and goose blood serum group.Conclusion1.The whole blood and serum of goose blood could inhibit the proliferation of human gastric cancer BGC-823 cells and induce its apoptosis,and the effect was similar,which indicated that the goose blood serum group contained effective components.2.The mechanism of inducing apoptosis in human gastric cancer BGC-823 cells by whole blood and serum of goose blood is related to the death receptor pathway. |
