1 Objective:Rapid identification of Radix Salviae Miltiorrhizae from four main production areas by three methods,to provide a scientific evidence for studying overall quality control and evaluation of Radix Salviae Miltiorrhizae.2 Methods:To identify the two kinds of Radix Salviae Miltiorrhizae and Radix Salvia bowleyana,and to distinguish Radix Salviae Miltiorrhizae from different origins,different parts by electrochemical fingerprint technology,infrared spectrum technology and Fourier-Transform Infrared Microspectroscopy.3 Results:3.1 The electrochemical fingerprints of Radix Salviae Miltiorrhizae were studied,the optimum conditions of the Belousov-Zhabotin-skii(B-Z)oscillation were as follows,the temperature of 310 K,the speed of 400 r/min and adding 0.2000 g of Radix Salviae Miltiorrhizae powder.The electrochemical fingerprints and Characteristic Parameters of Radix Salviae Miltiorrhizae from different origin,different parts and Radix Salvia bowleyana were obtained and discussed.3.2 The absorption peak and intensity of Infrared(IR)spectra of Radix Salviae Miltiorrhizae from different origins were significantly different,the content of tanshinones,salvianolic acid and carbohydrate in Radix Salviae Miltiorrhizae from Deyang and Luoyang were higher.The results were verified by second derivative infrared spectroscopy.The FTIR spectra of different parts of Radix Salviae Miltiorrhizae(the root head,main root,fibrous root)were different.The characteristic absorption peak of IR spectra of fibrous root at 1 026 cm-1was more obvious than others.It is suggested that the content of tanshinones,salvianolic acid and carbohydrate in the fibrous root is higher than that in the root head and main root.The IR spectra of Radix Salvia miltiorrhiza and Radix Salvia bowleyana were quite different,the characteristic absorption peaks of IR spectra at 1 610 cm-1,1 026 cm-1,the peak intensity of Radix Salvia miltiorrhiza was higher than Radix Salvia bowleyana,the absorption peak of second derivative infrared spectrum at 1 118 cm-1,the peak intensity of Radix Salvia miltiorrhiza was more obvious than Radix Salvia bowleyana.SPSS software was used to construct a dendrogram of Radix Salvia miltiorrhiza from different origins,cluster analysis showed that 10 samples were divided into three groups in the distance of 10.3.3 The FTIR images of Radix Salviae Miltiorrhizae were studied,According to the level of transmittance,the best conditions of Fourier Transform Infrared Microspectroscopy were determined,Detection mode of transmission,Ba F2windows,10 um thick sections of Radix Salviae Miltiorrhizae.The FTIR images of Radix Salviae Miltiorrhizae from four main production areas were quite different.The tanshinones in Radix Salviae Miltiorrhizae from Deyang are mainly distributed in the cortex.The tanshinones in Radix Salviae Miltiorrhizae from Luoyang are mainly distributed in the formation layer.The tanshinones in Radix Salviae Miltiorrhizae from Hefei are mainly distributed in the upper center.The tanshinones in Radix Salviae Miltiorrhizae from Bozhou are mainly distributed in the ray position.The chemical images of Radix Salviae Miltiorrhizae from Linyi and other areas are not the same,which are mainly distributed in the center and cortex.4 Conclusion:.1 The electrochemical fingerprints of Radix Salviae Miltiorrhizae from different origins showed significantly different characteristics.Rapid identification Radix Salviae Miltiorrhizae from different origins,different parts and Radix Salvia bowleyana through the electrochemical fingerprints and the information parameters of Radix Salviae Miltiorrhizae.A rapid,simple and economical method was developed for identification of geographical origin and evaluation of Radix Salviae Miltiorrhizae samples by the electrochemical fingerprints.4.2 Identification of Radix Salviae Miltiorrhizae by FTIR and Clustering analysis,not only provide the information of main chemical constituents in Radix Salviae Miltiorrhizae,but also classify and identify Radix Salviae Miltiorrhizae from different habitats.The results indicated that there was a certain correlation between the effective constituents of Radix Salviae Miltiorrhizae and the production areas.The FTIR method was proved to be feasible for identify rapidly Radix Salviae Miltiorrhizae from different origins.4.3 The chemical images of Radix Salviae Miltiorrhizae were quite different,The FTIR Microspectroscopy technology was not only classify and identify the regions of Radix Salviae Miltiorrhizae,but also they can provide semi-quantitative research and spatially distributed information about the chemical composition of samples.The method has advantages such as intuitive,accurate and high sensitive,it comprehensively analyzes the differences in the intrinsic quality of Radix Salviae Miltiorrhizae from different origins.
