Objective : According to the high recurrence and metastasis of osteosarcoma in clinical characteristics.From normal osteosarcoma HOS cells in exploring the isolation and identification methods of stem cells.and detect its biological characteristics,laying a theoretical basis for further research to cure osteosarcoma Methods: From the Chinese academy of sciences(Shanghai cell bank)to buy osteosarcoma HOS cells,proliferation,mass culture by limited dilution method for osteosarcoma HOS cells for stem cell separation,to obtain cells,respectively,using real-time fluorescent quantitative PCR and Western blot method to detect Nanog,SOX-2,OCT4 m RNA expression and protein expression.At the same time detection ability to self-renewal capacity of stem cells proliferation and migration.Results: Osteosarcoma HOS cell lines to extend and multiply.By limited dilution method monoclonal the HOS-M cells has a strong ability of proliferation and migration,HOS-M cell surface Nanog,SOX-2,OCT4 m RNA and protein expression levels were significantly higher than that of parental HOS cells.Cell proliferation experiment and scratches the results found HOS-M cells has a strong ability of proliferation and migration.Conclusion: Limited dilution method can be used for the separation of osteosarcoma HOS cell,The experiment of osteosarcoma cells with stem cell properties.
