| Objective To observe the treatment effect of the Corydalis extract on CCl4 induced liver tissue pathology and blood biochemical indexes in model rats,and to explore the mechanism of the key protein expression of PDGF signaling pathway in rat hepatocytes,to evaluate the preventive and therapeutic effects of Corydalis extract on hepatic fibrosis in rats,in order to provide experimental basis for the research and development of new drugs.Methods 1.With the continuous extraction of tetrahydropalmatine extraction;Determination of tetrahydropalmatine in Corydalis extract by HPLC.2.CCl4 intraperitoneal injection was used to establish the rat model of hepatic fibrosis.Silymarin was taken as the positive control,and the pharmacodynamics of Corydalis extract was evaluated.3.Using HE staining to observe the effect of Corydalis extract on liver histological changes of rat liver fibrosis.4.Using Masson staining to observe the effect of liver fibrosis of Corydalis extract on liver fibrosis in rats.5.The contents of AST and ALT in liver fibrosis rats were detected by colorimetry.6.The content changes of hyaluronic acid(HA),hydroxyproline(Hyp),type Ⅲcollagen(PC Ⅲ)and typeⅣcollagen(PCⅣ)in rats with hepatic fibrosis were determined by enzyme-linked immunosorbent assay(ELISA),tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)the level of influence.7.Effects of Corydalis extract on expression of α-SMA,TGF-β1 and PDGF-BB in rats with liver fibrosis by immunohistochemical method.Results Compared with the model group,Corydalis extract can significantly improve the pathological changes of liver tissue in rats with hepatic fibrosis;Significantly reduce serum ALT and AST,improve liver function;Significantly reduced serum HA,HYp,PC Ⅲ,PC Ⅳ,TNF-α and IL-1β levels,improve liver fibrosis levels;α-SMA in liver tissue was significantly decreased,which was mostly expressed in fibroplasia;The expression of PDGF-BB and TGF-β1 in the cytoplasm and membrane of the injured hepatocytes decreased significantly with the prolonged treatment time,which was mainly found in the portal area,which may be one of the mechanisms of the anti-hepatic fibrosis effect of the Corydalis extract.Corydalis extract significantly reduced the expression of PDGF-βr and p-p38 in the liver tissue of rats with hepatic fibrosis,suggesting that the anti-hepatic fibrosis effect may be through the activation of PDGF signal transduction pathway.Conclusion Corydalis extract can significantly improve the general status of liver fibrosis in rats,liver function and the degree of liver fibrosis,liver fibrosis have a better therapeutic effect.The mechanism may be the inhibition of PDGF-βr and p-p38 expression in liver tissue to inhibit the PDGF signal transduction pathway,in order to achieve anti-liver fibrosis. |
