Differential Expression Of AKT3 In Endometrium And Clinical Observations Of Patients With PCOS Combined With IR After The Treatment On Tanshinone

Research purpose:The goal of this research was to study the differential expression of AKT3 in endometrium of PCOS(polycystic ovary syndr)patients combined with IR(insulin resistanc)before and after tanshinone treatment by using PCR Technology.We could explore the changes of gonadotropin before and after tanshinone therapy,and provide new ideas for the clinical treatment of PCOS patients combined with IR.Methods:The cases of the recruited patients suffered from PCOS with IR in this study were selected from the National Center for Chinese Medicine Clinical Research Center sub-center Huai’an Maternal and Child Health Hospital.We had studied the changes of endometrial tissue and peripheral sera of the patients before and after they were treated with tanshinone capsules for three months.The differential expression of AKT3 gene in endometrial tissue before and after treatment was detected by qRT-PCR.So we could explore the target of tanshinone.The changes of serum androgen levels were measured by ELISA and electrochemiluminescence before and after treatment.The experimental data were analyzed by t test and statistically analyzed,so we could determine the statistical significance if P<0.05.Results:(1)The expression of related genes in endometrial tissue:the relative expression of AKT3 mRNA in endometrial tissue of PCOS patients combined with IR after they were treated with tanshinone was significantly decreased(P<0.05).(2)The levels of serum androgen:after the PCOS patients combined with IR were treated with tanshinone,the levels of serum androgen tended to be normal,and the levels of DHEAS and AND were significantly increased,and the SHBG level was significantly decreased(P<0.05).Conclusion:(1)During the treatment of PCOS patients combined with IR,tanshinone capsules can regulate the related genes in endometrial tissues.(2)Similarly,the treatment of tanshinone,has played a significant effect on regulation of serum androgen synthesis in patients.This research has definite clinical significance.Therefore,traditional Chinese medicine has a broader prospect and explore space in the prevention and treatment of PCOS combined with IR and its related endometrial lesions.
