| Objective: To investigate the effect of Qiweibaizhusan on bacteria lactase gene diversity of intestinal contents in dysbacteriotic diarrhea mice,and provided the basis for its curative effect mechanism from the angle of bacteria lactase.Methods: Thirty trialmice were randomly divided into normal group(n=12),model group(n=12)and Qiweibaizhusan group(n=6).All the mice were administrated with mixed antibiotic(62.5 g/L)by the dose of 0.4 m L for each one and each time to bulid dysbacteriosis diarrhea model other than the normal group.2 times a day,five consecutive days.After the model was established,the Qiweibaizhusan group was given Qiweibaizhusan decoction based on the dose of 0.16 g/(kg·d)to treatment,while the model and control group were given equal amount of distilled water,2 times a day,three consecutive days.After model and treatment,the contents of jejunum and ileum were extracted respectively,and then bacterial genomic DNA was extracted.The diversity analysis of bacterial lactase gene in intestinal contents was conducted by Miseq Metagenomic sequencing.Results: After the model of dysbacteriosis diarrhea,the 871 OTUs of model group was less than 963 OTUs of normal group,807 of them were the same.The Chao1 index and ACE index of normal group was higher than model group(P<0.05).,by the treatment of Qiweibaizhusan,the 884 OTUs of bacteria lactase gene in Qiweibaizhusan group was far below that 1056 OTUs in normal group or 1143 OTUs in model group.The Chao1 index,ACE index and Shannon index of Qiweibaizhusan group were all far below the normal group or model group,and there have significant statistical significance in Shannon index(P<0.01 or P<0.05).From the point of Beta diversity analysis,after the model of dysbacte riosis diarrhea,samples of normal group were relatively concentrated distribution,while the model group were relatively discreted distribution and the distance with the normal group were large,by the treatment Qiweibaizhusan,there had a certain distanc e between the normal group,model group or Qiweibaizhusan group each other.From the point of species evolution and abundance analysis,bacterial lactase gene in intestinal contents mainly from Actinobacteria,Proteobacteria,Firmicutes and most Unclassfied,Proteobacteria was dominant species in known species,the abundance of Bradyrhizobium sp.BTAi1,Agrobacteriumsp.H13-3,Acidovorax sp.KKS102,Azoarcus sp.KH32 C and Aeromonas caviae in model group were lower than normal group,while with the treatment of Qiweibaizhusan,the abundance of Acidovorax sp.KKs102 and Pseudomonas oleovoransin Qiweibaizhusan group were higher than normal group or model group.After the model of dysbacteriosis diarrhea,the normal group and model group had 79.33±0.58 and 76.00±4.58 different genera sources respectively,Gordonia,Mycobacterium,Frankia,Microbacterium,Novosphingobium,Aeromonasonly exist in normal group,Micromonospora,Paenibacillus,Ensiferonly exist in model group,the higher abundance levels of species in t he known species were Corynebacterium,Arthrobacter,Bradyrhizobium,Acidovorax,Azoarcus,Enterobacter,Klebsiella,Pseudomonas.The genus with high abundance level between normal group and model group separated apparently,the abundance of lactase gene of Gordonia,Frankia,Novosphingobium,Mycobacterium,Agrobacterium,Rhizobium,Escherichia,Acidovorax,Aeromonas,Microbacterium in the normal group was high.After the treatment of Qiweibaizhusan,the normal group,model group and Qiweibaizhusan group had 82.33±5.13,93.33±8.39 and 72.00±5.00 different genera sources respectively,and the Qiweibaizhusan group was significant lower than model group(P<0.01),Rhodococcus,Paenibacillus,Ensifer only exist in normal group,Ruminococcus,Methylarcula only exist in model group,the high abundance of lactase gene were Corynebacterium,Arthrobacter,Eggerthella,Acidovorax,Klebsiella,Pseudomonas.Qiweibaizhusan increased the lactase gene abundance of Stenotrophomonas,Eggerthella,Acidovoorax anddecreased the lactase gene abundance of Fretibacterium,Serratia,Novosphingobium,Rhizobium,Microbacterium,Methylarcula,Corynebacterium,Micromonospora of model mice.Conclusion: There was high diversity of bacterial lactase gene in intestinal contents.The diversity of bacterial lactase gene in intestinal contents decreased significantly after mice dysbacteriosis diarrhea induced by antibiotic,and the community structure of lactase gene was difference obviously compared with the normal group.It embogyied in decrea sing the abundance of Bradyrhizobium sp.BTAi1,Agrobacteriumsp.H13-3,Acidovorax sp.KKS102,Azoarcus sp.KH32 C,Aeromonas caviae,Gordonia,Frankia,Novosphingobium,Mycobacterium,Agrobacterium,Aeromonas,Microbacterium.After the treatment of Qiweibaizhusan on dysbacteriosis diarrhea,the diversity of bacterial lactase gene had not increased,it mainly changed the community structure of lactases gene.It embogyied in increasing the abundance of Eggerthella,Acidovoorax,Stenotrophomonas,and decreasing the abundance of Fretibacterium,Serratia,Novosphingobium,Rhizobium,Microbacterium,Methylarcula,Corynebacterium,Micromonospora in model mice. |
