Objective: Adiponectin,an adipose tissue-derived bioactive molecule,is widely distributed in various human tissues and cells.It plays an important role in the development of many kinds of tumors.This study aimed to explore the influence of different concentration of adiponectin on different growth of breast cancer cell lines in vitro and to investigate the distribution of adiponectin and adiponectin receptors in breast cancer patients with different clinical characteristics,and compare the difference between the expression of adiponectin in peripheral blood serum and tissues of breast cancer patients and normal women.Methods:1、According the concentration of adiponectin,it can be divided into the low concentration group(0.5μg/ml)and high concentration group(15μg/ml).Detect the influence of different breast cancer cell line,such as MCF-7,MDA-MB-231,SKBR-3 in two groups by MTT method.2、According to the 2012 WHO Classification of Tumors of the Breast,40 cases of breast cancer paraffin-embedded tissues treated in Tianjin Medical University Cancer Institute and Hospital from 2013 December to 2014 August,and 40 cases of normal adjacent tissues were collected.All patients underwent radical mastectomy or modified radical mastectomy or conservative surgery.The expression of adiponectin and adiponectin receptors were detected by immunohistochemistry Investigate the distribution of adiponectin and adiponectin receptors in the two groups,and analyze the correlation between them and different clinical characteristics.3、The peripheral blood serum samples of 100 cases of breast cancer and 10 cases of advanced breast cancer including local advanced and distant metastasis were collected in Tianjin Medical University Cancer Institute and Hospital from 2013 December to 2014 August.And 100 healthy volunteers from the physical examination center of Tianjin Medical University Cancer Institute and Hospital enrolled as healthy controls.The concentration of adiponectin was detected by enzyme linked immunosorbent assay(ELISA).Investigate the concentration of adiponectin in the two groups,and analyze the correlation between them and different clinical characteristics.Results:1、Compared with the control group,the influence of breast cancer cell growth in two groups is different.There was inhibition in high concentration group(15μg/ml)to some extent,and the inhibition rate were 50.4%,36.4% and 61.5% respectively.However,the influence of the low concentration group(0.5μg/ml)was not obvious.2、The expression of Adiponectin in breast cancer tissues and normal breast tissues were 57.5% and 92.5% respectively.Compared the controls,we found the expression of Adiponectin was lower,and the difference had statistical significant(P values < 0.001).The lower of expression of adiponcetin,the larger of tumor size,and the superior of histological grading was in breast cancer patients.And it showed negative associated with axillary lymph node metastasis.However,it showed not obviously correlation between it and age,length of the course of the disease,clinical stage,and the expression of ER/PR,and so on(P values > 0.05).3、The concentration of adiponectin in the peripheral blood serum samples of 100 cases of breast cancer was 0.22 ~14.83μg/ml,and the median concentration was 3.34μg/ml.It was 0.25~4.52μg/ml in 10 cases of advanced breast cancers,and the median concentration was 1.47μg/ml.In the normal controls,it was 1.20 ~24.86μg/ml,and the mean concentration was 9.05μg/ml.Compared to healthy people,the levels of adiponectin in the peripheral serum of breast cancer was lower significantly,and showed negative associated with the development of disease course.4、According to the normal female levels of adiponectin in the peripheral serum,100 patients with breast cancer were divided into low concentration group(<9.05μg/ml)and high concentration group(>9.05μg/ml).In 100 cases breast cancer patients,our study showed that the levels of adiponectin in the peripheral serum had negative associated with BMI,clinical stage,histological grading and axillary lymph node metastasis.Actually,we found that the levels of adiponectin at III stage were lower than 0-II stage(P values <0.001),histological III grade were lower than histological I-II grade(P values <0.05).The levels of adiponectin in patients with axillary lymph node metastasis were lower than the patients without lymph node metastasis(P values < 0.05).But,there was no significant statistical difference between it and the family history of cancer,tumor size,pathologic type,and the expression of ER/PR,C-erb B-2 and so on(P values > 0.05).Conclusion:1、Even though the influence of the low concentration adiponectin was not obvious,there was inhibition in high concentration group to some extent.2、Compared the normal breast tissues,the expression of Adiponectin was lower in breast cancer tissues,and showed negative associated with tumor size,histological grade and axillary lymph node metastasis.The difference had statistical significant(P values < 0.05).3、The levels of adiponectin in the peripheral serum of breast cancer was lower than healthy women,and showed negative associated with the development of disease course.4、The levels of adiponectin in the peripheral serum had negative associated with BMI,clinical stage,histological grading and axillary lymph node metastasis.The difference had statistical significant(P values < 0.05).
