Objective: To explore the KYNU(Kynureninase)protein′s influence on He La/DDP cell′s biological properties.Methods: 1.Cultivate He La cell and He La/DDP cell.Use the q PCR and Western Blot methods to respectively test the expression of KYNU m RNA and KYNU protein in He La cells and He La/DDP cells.2.Construct the plasmid which can overexpress the KYNU,enhance the expression of KYNU in cells which lower express KYNU protein.3.Application of Transwell method and flow cytometry to respectively test the changes of Cell migration,cell invasion ability and Cell apoptosis rate after the KYNU protein overexpression.Results: 1.It is find that the expression of KYNU m RNA and KYNU protein in He La cells are higher than He La/DDP cells after using the q PCR and Western Blot methods.2.Transwell method is applied to detection the cell migration ability of He La/DDP cells,He La/DDP-empty cells and He La/DDP-KYNU cells,it is found that the number of He La/DDPgroup,He La/DDP-empty group and He La/DDP-KYNU group that across the transwell chambers after 24 hours are 125±6,123±5 and 68±5respectively.The cell migration ability of He La/DDP-KYNU cells is lower than He La/DDP-empty cells(P﹤0.05).Transwell method is applied to detection the cell invasion ability of He La/DDP cells,He La/DDP-empty cells and He La/DDP-KYNU cells,it is found that he number of He La/DDP group,He La/DDP-empty group and He La/DDP-KYNU group that across the transwell chambers after 24 hours are69±9,67±5 and 65±4 respectively.KYNU protein not influence the cell invasion ability of He La/DDP cells(P>0.05).5.The cell apoptosis rates of He La/DDP group,He La/DDP-empty gpoup and He La/DDP–KYNU group are3.2%±0.09%,4.0%±1.9%and 32.4%±3.3% respectively.The rates of cell apoptosis in He La/DDP – KYNU cells is higher than He La/DDP-empty cells(P﹤0.05).Conclusions: 1.The lower expression of KYNU m RNA and KYNU protein in He La cells indicate that the lower expression of KYNU may influence the He La cells resistant cisplatin.2.KYNU inhibit Hela/DDP cells migration ability and enhance their apoptosis in vitro.3.KYNU expression can help reduce Hela/DDP cell ′s migration ability and promote cell apoptosis which is expected to effectively become the therapeutic target of cervical adenocarcinoma that resistant cisplatin. |
