Effect Of Yougui Drink(右归饮) On Articular Cartilage Degeneration In Rats Model With Osteoarthritis Via Regulating Expression Of Stromal Cell Derived Factor-1

Objective This experiment was performed to study the effect of Yougui drink(右归饮)on articular cartilage degeneration in rats model with osteoarthritis(OA)via regulating the expression of SDF-1.Methods 30 male SD rats were randomly divided into saline group,prevention group and treatment group,created into OA model,and were intragastric administrated by equivalent yougui drink and saline respectively.8 weeks later,all the 30 rats were sacrificed,their blood was collected and the level of SDF-1,interleukin-1β(IL-1β),tumor necrosis-α(TNF-α),matrix metalloprotease-13(MMP-13)were quantified.Knee joints and articular cartilages from 3 groups were obtained to be analyzed by micro-CT in the differences of microstructure and bone histomorphometry.Besides,aggrecan mRNA and collagen type Ⅱ protein levels in cartilage tissues were measured respectively.Statistics were peformed by using SPSS 17.0 software.Results Data of serum index,aggrecan mRNA and collagen type Ⅱ protein levels in prevention group were lower than that of other 2 groups(P<0.01),there was no significant difference between treatment group and saline group(P>0.05).Imaging observation reveals obvious articular cartilage degeneration in saline and treatment group.In prevention group,there was a significant decrease in osteoproliferation.In prevention group,statistics of trabecular separation(Tb.Sp),trabecular number(Tb.N),the percentage of the relative volume of trabecular(BV/TV)and bone mineral densiy(BMD)are different with the other 2 groups(P<0.01&P<0.05),and the difference between saline and treatment group are unsignificant(P>0.05).Conclusions Yougui drink could inhibit the expression of SDF-1,IL-1β,TNF-α in the whole process of OA as well as attenuate articular cartilage degeneration at the early stage.
