Effects Of Yi Qi Bu Shen Fang On The Balance Of Th17/Treg And The Related Cytokines Of Peripheral Blood In Abortion-prone Mice

Objective To observe the adjustment of Chinese medicine with the function of Yi Qi Bu Shen Fang on the balance of Th17/Treg,the related cytokines in abortion-prone mice peripheral blood and the pregnancy outcomes,discuss the mode of action of Yi Qi Bu Shen Fang adjust maternal-fetal immune,which can offer a new theoretical foundation for Yi Qi Bu Shen Fang cure recurrent spontaneous abortion.Methods The CBA/J females were mated with BALB/c male mice,which is comformd as the normal control group;CBA/J female mice were mated with DBA/2 male mice to establish abortion-prone model.The CBA/J pregnant rats of experimental group were randomly divided into 5 groups:the high dose of Chinese medicine group,the medium dose of Chinese medicine group,the low dose of Chinese medicine group,the negative control group,the positive control group.Mice were sacrificed on the 14th day of gestation,the embryo absorption rate was counted and their peripheral blood was taken out.Analtsis the expression of CD4+IL-17+cell.CD4+CD25+Foxp3+celland ratio with Flow cytometry analysis technique(FCM),and analysis the protein content of related cytokines(IFN-γ、IL-17、IL-21、IL-4、IL-10)by Enzyme-linked immunosorbent assay.Results(1)Embryo Absorption Rate:Embryo Absorption Rate arranged from high to low is the negative control group、the low dose of Chinese medicine group、the high dose of Chinese medicine group、the positive control group、the medium dose of Chinese medicine group、the normal control group.Compared with the negative control group,embryo absorption rates of all the therapeutic groups were reduced(P<0.05),but no significant differences between them(P>0.05).(2)The expression tendency of CD4+IL-17+T cells arranged from high to low is the negative control group、the normal control group、the low dose of Chinese medicine group、the medium dose of Chinese medicine group、the high dose of Chinese medicine group、the positive control group.Differences between the negative control group and the medium dose of Chinese medicine group、the high dose of Chinese medicine group、the positive control group has a statistical significance(P<0.05).(3)The expression tendency of CD4+CD25+Foxp3+Tcells arranged from high to low is the positive control group、the low dose of Chinese medicine group、the medium dose of Chinese medicine group、the high dose of Chinese medicine group、the normal control group、the negative control group;Comparing with the negative control group,expression of CD4+CD25+Foxp3+T cells of the low dose of Chinese medicine group、the medium dose of Chinese medicine group、the positive control group increased,which has a statistical significance(P<0.05).(4)The proportion of the CD4+IL17+and CD4+CD25+Foxp3+T cells arranged from high to low is the negative control group、the normal control group、the low dose of Chinese medicine group、the high dose of Chinese medicine group、the medium dose of Chinese medicine group、the positive control group.Comparing with the negative control group,The proportion of the positive control group and all the therapeutic groups of chinese medicine reduced,which has a statistical significance(P<0.05).(5)Cytokines:in peripheral blood,Comparing with the negative control group,The expressions of IFN-y、IL-17、IL-21 were reduced while the expressions of IL-4、IL-10 were increased among all the therapeutic groups.The expressions of IFN-γ、IL-17 of the low dose of Chinese medicine group reduced(P<0.05),while the expressions of IL-10 increased(P<0.05).The expressions of IFN-γ、IL-17 of the medium dose of Chinese medicine group reduced(P<0.05),while the expressions of IL-4 increased(P<0.05).The expressions of IFN-γ、IL-17、IL-21 of the high dose of Chinese medicine group reduced(P<0.05).There is no significant differences between all the chinese medicine groups except the expressions of IL-17 between the low and medium dose of Chinese medicine groups.Conclusion ①Chinese Medicine Yi Qi Bu Shen Fang can reduce the embryo absorption rates of establish abortion-prone model mices;②Chinese Medicine Yi Qi Bu Shen Fang can reduce the expression of CD4+IL-17+cells and increased CD4+CD25+Foxp3+ cells of the abortion-prone mice;③Chinese Medicine Yi Qi Bu Shen Fang can reduce the proportion of the CD4+IL17+and CD4+CD25+Foxp3+T cells;④)Chinese Medicine Yi Qi Bu Shen Fang can also reduce the expression of IFN-γ、IL-17、IL-21 and increase the the expression of IL-4、IL-10;⑤There is a probability that the chinese medicine can adjust the expression tendency of Th17 cells and Treg cells,reduce the proportion of the Thl7 and Treg cells,meanwhile,it can also adjust the expression tendency of cytokines,influence the balance of Th17 and Treg cells,which leading to the immunological tolerance,thus improving the pregnancy outcome.
