| Objective:To explore the effect of ginsenoside Rbl(GRb1)on aplastic anemia mice hematopoietic and on dendritic cells(DCs)differentiation.Methods:BALB/C mice model of immunemediated aplastic anemia was established by radiation with sublerhal dose of 60Coγ following the intravenously infusing lymphocytes of DBA/2 mice.The experiment was randomly divided into 5 groups,including normal control group,AA model group,GRb1 treatment groups with low(10 mg/Kg),medium(20 mg/Kg)and high(40 mg/Kg)doses,and positive control group with cyclosporine A(CSA)respectively by intraperitoneal injection per day.Model mice in the control group and normal mice in the normal control group were treated with normal saline.We killed all the mice at 12th and detected the peripheral hemogram,reticulocyte count(Ret),bone marrow stem cell count and T lymphocyte subsets and DCs molecules on the surface level of every groups.Results:Compared with normal group,model group mice peripheral white blood cells(WBC),hemoglobin(Hb)and platelets(PLT),Ret,bone marrow stem cells count,auxiliary T lymphocytes(CD3+,CD4+)levels,helper T lymphocytes and inhibitory ratio of T lymphocytes(CD4+/CD8+)were significantly reduced(P<0.05),weak T lymphocytes(CD3+CD8+),peripheral blood DCs surface molecule CD86 significantly increased(P<0.01).Verifiable detection indicators comply with the AA model group mice.Compared with model group:(1)GRb1 low,medium and high dose group,the mice the WBC CSA group were significantly increased(P<0.05);(2)GRb1 Hb,high dose group,the CSA group mice PLT count increased significantly(P<0.05);(3)GRb1 low,medium and high dose group,the CSA group mice CD34+were significantly increased(P<0.05).(4)GRb1 low,medium and high dose group,the CSA group proportion of CD3+CD8+T cells in mice significantly reduced,the proportion of CD3+CD4+T cells,CD4+1 CD8+ratio were significantly increased(P<0.05);(5)GRb1 CD86+high dose group,the CSA group mice were significantly reduced(P<0.01).Compared with CSA group:(1)GRb1 low,medium and high dose group were no significant differences of the WBC,Hb,Ret(P st13 0.05);(2)GRb1 PLT count low dose group mice significantly decreased(P<0.05),GRb1 PLT count high,middle dose group mice without obvious difference(P st13 0.05);(3)GRb1 low,medium and high dose group mice CD34+ratio has no obvious difference(P st13 0.05);(4)GRb1 low,medium and high dose group of CD3+ CD8+T cells in mice proportion,the proportion of CD3+CD4+T cells,CD4+/CD8+ ratio has no obvious difference(P st13 0.05);(5)GRb1 CD86+low dose group mice were significantly increased(P<0.05),GRb1 CD86+levels of high,middle dose group mice have no obvious difference(P st13 0.05).Conclusion:proper dosage GRb1 type can promote immune mediated AA mice hematopoietic function,improve the level of peripheral blood cells and bone marrow stem cells count,has therapeutic effect on AA,curative effect is similar to the CSA without the side effects of larger,its mechanism may be related to inhibition of mature dendritic cells and regulation of CD4+/CD8+T cells,T cell percentage.Its role in treating AA is worth further research and discus... |
