| Objective The models of chronic heart failure in rats were established by abdominal aorta constriction,and randomly divided into model group,digoxin group,perindopril group,semen lepidii water extract group,semen lepidii alcohol extraction liquid group,plus sham group(which were separated abdominal aorta but not ligation),analyze the effects of different interventions on myocardial apoptosis and hemodynamics in rats with chronic heart failure.Further clarify the semen lepidii water extract and alcohol extract on rats about heart failure survival rate,cardiac function,myocardial systolic and diastolic function,raise awareness of treatment of heart failure of effective components of semen Lepidii.In addition,this research improve understanding of treatment about traditional Chinese medicine anti heart failure,application of expanding Chinese medicine in the treatment of heart failure.Methods 90 male clean grade SD rats were enrolled,weight 200±20 g,80 of them by abdominal aortic constriction method for chronic heart failure rat model,the remaining 10 were used as sham group.The rats is successfully established,which were randomly divided into model group,digoxin group,perindopril group,semen lepidii water extract group,semen lepidii alcohol extraction liquid.The structure and function of the heart were detected by ultrasonic cardiogram and sixteen-channel physiological recorder after 8 weeks of continuous administration,and cardiac hypertrophy index was calculated;HE and Masson staining were used to observe the pathological changes of myocardial tissue and structure,And comprehensive evaluation of semen lepidii seu extract of chronic heart failure rats and hemodynamic efects of myocardial cell apoptosis.All the data were analyzed using SPSS 17.0 statistical software for statistical analysis.Results1.Ultrasonic cardiogram detection:Compared with the control group,digoxin group,perindopril group,semen lepidii water extract group,semen lepidii alcohol extraction liquid group of interventricular septum(IVS)and left ventricular wall thickness(LVPW),left ventricular diastolic diameter(LVIDd)and systolic diameter(LVIDs)significantly decreased;left ventricular ejection ejection fraction(LVEF)and minor axis shortening(FS)increased significantly.Compared with digoxin group,semen lepidii water extract group,perindopril group,semen lepidii alcohol extraction liquid had statistical difference on improving ventricular remodeling,it had no statistical difference.There were statistical differences between the model group and the sham operation group.2.Hemodynamic detection:Compared with the model group,digoxin group,perindopril group,semen lepidii water extract group,semen lepidii alcohol extraction liquid heart rate(HR)and left ventricular end diastolic pressure(LVEDP)significantly decreased;left ventricular systolic pressure(LVSP),left ventricular pressure maximum rate of rise(+DP/dtmax)increased significantly;left ventricular pressure decline rate(-DP/dtmax)absolute value increased significantly.Compared with digoxin group,perindopril group,semen lepidii water extract group,semen lepidii alcohol extraction liquid group+dP/dtmax slightly increased,dP/dtmax slightly lower,there are no statistical difference in indicators.There were statistical differences between the model control group and the sham operation group.4.Detection of myocardial tissue pathology:Compared with the model group,sham group,digoxin group,perindopril group,semen lepidii water extract group,semen lepidii alcohol extraction liquid group myocardial cell apoptosis rate were significantly decreased.Conclusion This study have shown that semen lepidii water extract group,semen lepidii alcohol extraction liquid group can effectively improve heart function,reduce myocardial fibrosis.In improving hemodynamics,semen lepidii alcohol extraction liquid group was better than semen lepidii water extract group. |
