Objective To study alkaloid contents,inorganic elements contents and fingerprint in Fritillaria thunbergii Miq.from 34 different growing areas all over the china,in order to provide experimental evidence for quality control;Discuss the influence law of physiological characteristics,alkaloid contents and biomass under the different light intensity;To study the quality suitability rank dividing of Fritillaria thunbergii Miq.on the basis of investigation on the correlation between the ratio of Peimine and Peiminine in Fritillaria thunbergii Miq.and ecological factors,in order to provide scientific basis for its planting area of high-quality medicinal materials.Method 1.To determine the content of Peimine,Peiminine and Peimisine in Fritillaria thunbergii Mig.from different growing areas with HPLC-ELSD.2.Using similarity analysis,principal component analysis and cluster analysis to determine its total fingerprint pattern.3.ICP-OES method was used for the determination of nine inorganic elements,analyzing their correlation between the active ingredient and inorganic elements.4.Adopt plastic net shade,four different kinds of light intensities were set,with the light transmittance were controlled group CK(100%)、T1(50%)、T2(25%)、T3(5%)respectively,photosynthesis parameters,alkaloid content and biomass in different kinds of shading were detected respectively.5.Based on the samples quality of analysis 34 areas of Fritillaria thunbergii Miq.,relational model was constructed Peimine,Peiminine and environmental factors,the use of ArcGIS Spatial computing was applied to assess the quality suitability rank dividing Fritillaria thunbergii Miq.in china.Result 1.The three constituents showed good linearity(R2≥0.9993)in the range of the tested constituents.The average recoveries(n=6)of the method were 99.11%、98.11%and 99.38%,respectively.The contents of the three constituents in Fritillaria thunbergii Miq.from thirty-four different growing areas were 0.02%-0.18%、0.01%-0.07%and 0.01%-0.04%,respectively.2.An HPLC-ELSD fingerprint was developed for alkaloid in Fritillaria thunbergii Miq.quality evaluation.The results showed that alkaloid in Fritillaria thunbergii Miq.had been effectively separated,and 7 common characteristic peaks,identified as characteristic fingerprint peaks in HPLC-ELSD fingerprint were found in all the samples.Samples from different regions were of high similarity,which were in consistent with the results of the cluster analysis and principal component analysis.3.Determination of 34 habitat Fritillaria thunbergii Miq.samples of 9 kinds of inorganic elements,the content of inorganic elements in Fritillaria thunbergii Miq.from 34 different growing areas were different;as,As,Hg,Cr content of heavy metals exceeded.The result of correlation analysis showed that Peimine contents of Fritillaria thunbergii Miq.have strong a positive correlation with the Fe contents;Fe contents showed a positive correlation with Ca、Cu contents,Zn contents showed a positive correlation with Ca、Cu、Fe contents.4.Shading has the most significantly.impact on photosynthesis alkaloid contents and biomass.The moderate shade can increased net photosynthetic rate.The yield of Fritillaria thunbergii Miq.was lower when the light transmittance was in 5%.5.Higher alkaloid content in Fritillaria thunbergii Miq.is concentrated in the two provinces of Zhejiang and Jiangsu,which coincides with the production of practical experience,can provide scientific basis for the standardization of Fritillaria thunbergii Miq.cultivation and introduction.Conclusion 1.Fritillaria thunbergii Miq.on different growing areas of origin,the content of Peimine,Peiminine and Peimisine were determined to establish a multi-index components medicinal Fritillaria thunbergii Miq.Quality control system to ensure the quality of the Fritillaria thunbergii Miq.Provided.2.The establishments of a fingerprint quality evaluation system of Fritillaria thunbergii Miq.,can fully evaluate the quality of Fritillaria thunbergii Miq.3.The inorganic elements of Fritillaria thunbergii Miq.can provide of its identification,cultivation,application.4.75%shade conditions,help increase the net photosynthetic rate Fritillaria thunbergii Miq.,beneficial to photosynthesis.95%shading condition,seriously affecting the accumulation biomass.Shading and alkaloids between the two are not completely parallel.5.Fritillaria thunbergii Miq.optimal planting base area are mainly distributed in Zhejiang province,eastern Jiangsu province and other places.
