| Objective:Synovial sarcoma(SS)is a mesenchymal malignant tumour that exhibits epithelial differentiation by both morphological and immunohistochemical criteria.To investigate the application feasibility of the LCM method and tissue chip separation method in SS of formalin-fixed,paraffin-embedded(FFPE),and detect the expression of EMT/MET-related genes in spindle cells and epithelial cells of SS for strong technical support on the sutdy in the biphsic differentiation of SS.Methods:10 cases of FFPE primary SS,and 2 recurrence cases were collected for study,including 10cases of biphasic synovial sarcomas(BSSs),and 2 cases of monophasic fibrous synovial sarcomas(MFSSs).Arcturus XT~?system Laser capture microdissection(LCM)and tissue chip separation method were used for separation and purification of SS to the spindle cells and epithelial cells.EMT-related genes(E-cadherin,N-cadherin,Snail and Slug)were tested by q RT-PCR in spindle cells and epithelial cells.The SPSS17.0 Software was used for statistical analysis,by the control sample T test to compare the different between the groups.Results:(1)Capturing more than 2×10~4 of synovial sarcoma cells by using the Arcturus XT~?system LCM,which were used to extract RNA for Real-Time PCR,the success rate is 75%(3/4).(2)Tissue chip separation method can be used to separate paraffin synovial sarcoma,the success rate of RNA extraction was 72.2%(13/18).(3)The expression of m RNA in the samples from 11 cases of spindle cell of E-cadherin and N-cadherin are respectively 3.2116,2.5474;Snail and Slug are 22.5389,0.4985;in 5 cases of epithelioid cells,E-cadherin and N-cadherin are 2.9253,2.5628;Snail and Slug are 8.5432,0.4089.Of which,m RNA expression of Snail in spindle cell was significantly higher than that of epithelial area(P=0.001).(4)The expression of E-cadherin,N-cadherin,Snail and Slug of two primary synovial sarcomas in spindle area was significantly higher than that of recurrence(P values were 0.001,0.035,0.033,0.022).Conclusion:(1)The difference was found in epithelial and mesenchymal components of EMT-related gene,Snail in spindle cells was significantly higher than that of epithelial area in synovial sarcoma.(2)Both Arcturus XT?system LCM and Tissue chip separation method can be successfully used for Formalin-fixed,Paraffin-embedded tissue to separation and purification.Arcturus XT?system LCM can obtain the highly purified target cells. |
