Objects:To detect EGFR ex on19and exon21mutations status in peripheral blood of pre-and post-chemotherapy from advanced non-small cell lung cancer patients.To observe the impact of chemotherapy on EGFR mutation status. To provides some references for the application of EGFR-TKI in the treatment of lung cancer.Methods:Collect patients’ peripheral blood,extract the DNA in the plasma,analysis the DNA concentration,to applicate PCR for EGFR exonl9and exon21of plasma DNA after specific amplification,to use denaturing high-performance liquid chromatography to detect the exonl9and21mutation status.Results:In60patients,EGFR mutations were detected in30.0%(18/60) and25.0%(15/60) peripheral blood of pre-and post-chemotherapy.The EGFR mutation status was consistent in78.3%(47/60) patients. Among13discordant cases,8changed from pre-chemo mutant-type to post-chemo wild-type status,while5from pre-chemo wild-type to post-chemo mutant-type status.Conclusion:Chemotherapy may have influence on the peripheral blood EGFR mutation status in advanced non-small cell lung cancer patients.
