Effects Of Magnesium Supplementation On Chronic Kidney Disease-mineral And Bone Disorder In Rats With 5/6 Nephrectomy

Objective:The purpose of this study was to explore the effects of different concentrations of magnesium ion supplementation on chronic kidney disease mineral and bone disorder(CKD-MBD)in 5/6 nephrectomy rats,to provide a theoretical basis for delaying the progression of chronic kidney disease(CKD)and treating its complications.Method:SD rats were randomly divided into five groups: control group,model group,and administration group(low,medium,and high concentration magnesium group),with 10 rats in each group.The rats in the control group underwent the sham operation,and the model group and administration group were established by 5/6 nephrectomy.Magnesium ions were supplemented by gavage.The concentrations of the administration group were 12.5mg/kg/d,25 mg/kg/d and 50 mg/kg/d respectively.The experimental period was 12 weeks.Bone tetracycline was labeled on the 7th and 14 th days before the end of the experiment.The general state of rats was recorded on the day after operation,4 weeks,8 weeks and 12 weeks after operation.Serum samples of rats were collected to detect the levels of serum creatinine(Scr),blood urea nitrogen(BUN),magnesium,calcium and phosphorus.At the end of the experiment,the tissues of rat kidney and thoracic aorta were collected for hematoxylin-eosin(HE)staining,and the histopathological changes were observed under optical microscope.The left femur of rats was collected,the undecalcified bone sections were made,and the dynamic parameters were observed under the fluorescence microscope.Results:1.There was no significant change in the levels of Scr and BUN in the control group.Compared with the control group in the same period,the levels of Scr and BUN in the model group,low,medium and high concentration magnesium group increased(P<0.01).Compared with the model group in the same period,the levels of Scr and BUN in the low,medium and high concentration magnesium group decreased at 4,8,and 12 weeks(P<0.01).Compared with the low concentration magnesium group in the same period,the Scr level in the medium and high concentration magnesium groups increased at the 12 th week(P<0.05).2.There was no significant change in serum calcium level in the control group.Compared with the control group in the same period,the serum calcium level in the model group decreased(P<0.05),and the serum calcium level in the low,medium and high concentration magnesium group increased(P<0.05).Compared with the model group,the serum calcium level of rats in the low,medium and high concentration magnesium group increased at 4,8,and 12 weeks(P<0.05).Compared with the low concentration magnesium group in the same period,the serum calcium level in the medium and high concentration magnesium groups increased at the 12 th week(P<0.05).3.Compared with the control group,the serum phosphorus level of model group,low,medium and high concentration magnesium group increased gradually(P<0.05).The level of serum phosphorus in low,medium and high concentration magnesium group was lower than that in the model group at 4,8 and 12 weeks(P<0.05).Compared with the low concentration magnesium group in the same period,the serum phosphorus level in the high concentration magnesium group increased in the 4th week(P<0.05).4.A small amount of inflammatory cell infiltration was observed in glomerulus and stroma of rats in the control group.The renal tissue of the model group showed glomerulosclerosis,obvious proliferation of mesangial cells,necrosis and atrophy of renal tubules,protein tube type,and a large number of inflammatory cell infiltration in renal interstitium and glomerulus.The pathological changes of kidney tissue in low and medium concentration magnesium group were significantly better than those in the model group,and a large number of inflammatory cells could be seen in kidney tissue in high concentration magnesium group.5.There was no intimal thickening and arterial calcification in the control group.Calcification of thoracic aorta in model group.There was no obvious calcification in thoracic aorta of rats in low,medium and high concentration magnesium group.The intima of thoracic aorta in low and high concentration magnesium group was thickened.6.Compared with the control group,the growth rate of the femur in the model group decreased(P<0.05).Compared with the model group,the growth rate of the femur in the low,medium and high concentration magnesium group increased(P<0.05).Conclusions:1.Magnesium ions can increase serum calcium and decrease phosphate level in 5/6 nephrectomy rats.2.Magnesium ion can reduce vascular calcification and promote bone growth in 5/6 nephrectomy rats.3.Magnesium ions can reduce renal inflammation,protect renal function and delay the process of renal pathological changes in 5/6 nephrectomy rats.
