| Objective: To observe the effects of 10-week aerobic exercise on the level of S1 P and expression of Sph K1,S1 PL,TRAF2,S1PR1 and TNF-α in epididymal adipose tissue of obese rats,to explore the effect of aerobic exercise on Sph K1/S1 P inflammatory signaling pathway in adipose tissue of obese rats,and to improve the inflammatory mechanism of adipose tissue of obese rats.To provide experimental basis for the prevention and treatment of obesity tissue inflammation.Methods: Seventy-two 6-week-old SPF Sprague-Dawley male rats were randomly divided into 2 groups after 1 week of acclimation,and 8 rats in the control group were fed ordinary diet for 10 weeks.High fat diet obesity induction group(64)was fed high fat diet.By high fat feed rats after 10 weeks,weight more than 1.96 times the standard deviation of the normal control group the average weight of rats rats as building successful obese rats,which conforms to the condition of obesity rats 30,weight of the selected 16 as obesity experiment in rats,which were randomly divided into 2 groups: Obesity group(8)and Obese exercise group(8).The normal control group and the obese group did not exercise.The obese exercise group received moderate intensity exercise 5 times/week,60 minutes/time for 10 weeks.The first week was adaptive exercise.24 hours after the last platform exercise,rats in each group were sacrificed by cervical dislocation,and the samples were collected and preserved.The level of S1 P in epididymal adipose tissue was detected by ELISA.The expression of TNF-α protein in epididymal adipose tissue of SPHK1,S1 PL,TRAF2 and S1PR1 was detected by Western Blot.Results:(1)Epididymal fat volume and epididymal fat coefficient in obese exercise group were significantly lower than those in obese group(P<0.05).(2)The level of S1 P in epididymal adipose tissue in obese group was significantly higher than that in normal control group(9.02±0.56 VS 8.18±0.67,P<0.05);The level of S1 P in obese exercise group was significantly lower than that in obese group(8.35±0.53 VS9.02±0.56,P<0.05).(3)The expression of Sph K1 protein in epididymal adipose tissue of obese rats was significantly higher than that in normal control group(0.92±0.31 VS 0.49±0.29,P<0.05);The expression of Sph K1 protein in obese exercise group was significantly lower than that in obese group(0.60±0.21 VS 0.92±0.31,P<0.05).(4)There was no significant difference in the expression of S1 PL protein in epididymal adipose tissue between the obese group and the normal control group(0.86±0.35vs1.09±0.18,P>0.05);S1PL protein expression in obese exercise group was significantly higher than that in obese group(1.24±0.35 vs 0.86±0.35,P <0.05).(5)The expression of TRAF2 protein in epididymal adipose tissue of obese rats was not significantly different from that of normal control group(1.00±0.18 vs0.89±0.53,P>0.05);TRAF2 protein expression in obese exercise group was significantly higher than that in control group(1.40±0.29 vs 0.89±0.53,P<0.05),Significantly higher than the obese group(1.40±0.29 vs 1.00±0.18,P<0.01).(6)The expression of S1PR1 protein in epididymal adipose tissue of rats in obesity group was significantly higher than that in normal control group(1.72±0.38 vs1.04±0.20,P<0.01);The expression of S1PR1 protein in obese exercise group was significantly higher than that in normal control group(1.49±0.4 vs 1.04±0.20,P<0.05).(7)The expression of TNF-α protein in epididymal adipose tissue of obese rats was significantly higher than that of normal control group(1.47±0.22 vs 0.96±0.31,P<0.01);The expression of TNF-α in obese exercise group was significantly lower than that in obese group(1.18±0.14 vs 1.49±0.24,P<0.05).Conclusion:(1)The expression of Sph K1 and S1PR1 protein and the level of S1 P in epididymal adipose tissue of obese rats increased,which promoted the inflammation of adipose tissue.(2)Aerobic exercise can down-regulate Sph K1/S1 P inflammatory signal and improve adipose tissue inflammation in obese rats. |
