Objective:The Kunming mouse model of T2 DM was established by injecting streptozotocin(STZ)with high-fat diet for four weeks.The effect of intermittent fasting combined with aerobic exercise on T2 DM Kunming mice was observed,and the effect of intermittent fasting combined with aerobic exercise on T2 DM was analyzed.Influence of glucose and lipid metabolism in Kunming mice,so as to explore more suitable intervention methods for glucose and lipid metabolism in diabetes.Methods:Forty SPF 4-6 week-old Kunming male mice cultivated by Xinjiang Medical University were purchased.The mice were divided into 5 groups by random grouping,with 8 mice in each group.The blank control group(group C)was fed with normal feeding,and the other four groups of mice were fed with high-fat diet.Four weeks later,the high-fat-fed mice were injected with streptozotocin(STZ)intraperitoneally to induce T2 DM,and one week later,the fasting blood glucose of the mice was detected to determine whether the diabetic mouse model was successfully constructed.After successful modeling,the high-fat group was randomly divided into a diabetes control group(DC group,n=8),a intermittent fasting group(IF group,n=8),and a low-intensity exercise + intermittent fasting group(LI+IF,n=8).8),moderate intensity exercise + intermittent fasting group(MI+IF,n=8).The intermittent fasting group stopped eating after 19:00 Beijing time every day,and was given free water;the two groups of aerobic exercise combined with intermittent fasting performed aerobic exercise of different intensities while fasting,and were given free water.Mice body weights were measured 6 weeks after the intervention.In this study,biochemical detection instruments were used to detect glucose metabolism indicators in mice: blood glucose,glucose tolerance;lipid metabolism indicators: TC,TG,HDL-C,LDL-C.At the same time,the mouse liver was sliced and its cell morphology was observed.Finally,the transcription levels of glucose metabolism-specific genes: G6 P,PGC,PEPCK and lipid metabolism-specific genes:SREBP-1c,FAS,C/EBPα,PPARγ in mice were detected by fluorescence quantitative method.:SREBP-1c,C/EBPα,PPARγ,G6 P,PEPCK expression levels were detected.Results:(1)The body weight,average blood glucose concentration and glucose tolerance indexes of the mice in the intervention group were significantly reduced,and the reduction degree of the mice in the IF group was not as high as that of the mice in the intermittent fasting combined with aerobic exercise group.(2)The results of triglyceride,total cholesterol,high-density lipoprotein and low-density lipoprotein in the intervention group were significantly improved,and the intermittent fasting combined with aerobic exercise intervention group was better than the IF group.(3)The arrangement of hepatocytes in the DC group was disordered,the volume increased,and the hepatic sinusoids were obviously dilated.Compared with the liver tissue of the mice in the DC group,the liver tissue of the mice in the IF group,the LI+IF group,and the MI+IF group group had different degrees of improvement,and the liver tissue of the MI+IF group was the best,followed by the LI.+IF group.(4)The expressions of glucose metabolism-specific genes G6 P,PGC and PEPCK were significantly decreased in the intervention group.The LI+IF group had a stronger effect on the expression of related genes.(5)The expressions of lipid metabolism-specific genes SREBP-1c,FAS,C/EBPα,and PPARγ in the intervention group were significantly decreased,and the LI+IF group had a stronger effect on the expression of related genes.(6)The expressions of glucose and lipid metabolism-related proteins SREBP-1c,C/EBPα,PPARγ,G6 P,and PEPCK in the intervention group were decreased,and there were significant differences between the groups,and the MI+IF group had the best effect.Conclusion: Both intervention groups could improve glucose and lipid metabolism,and the effect of intermittent fasting combined with aerobic exercise was better.Intermittent fasting combined with aerobic exercise corrected the abnormal glucose and lipid metabolism in mice by improving the abnormal expression of genes and proteins related to glucose and lipid metabolism in the liver of mice,thereby improving the insulin resistance and glucose tolerance of mice.The possible mechanisms are as follows:(1)Intermittent fasting combined with aerobic exercise reduces the expression of SREBP-1c,thereby reducing the expression of C/EBPα and PPARγ,thereby slowing down the synthesis of fat.(2)Intermittent fasting combined with aerobic exercise reduces the expression of FAS by down-regulating the gene expression of SREBP-1c,thereby reducing the synthesis of fatty acids.(3)Intermittent fasting combined with aerobic exercise down-regulates the gene expression of PEPCK,which in turn reduces the expression of G6 P,thereby intervening in the process of gluconeogenesis in mice and reducing blood sugar.The comparison of multiple groups of data found that the MI+IF group had the best effect on improving the glucose and lipid metabolism in mice.
