ObjectiveIn this clinical trials,the levels of Hotair、lncRNA-p21、HCP5 and RP1-68D18.2 in two groups of patients,namely patients with rheumatoid arthritis(RA)and healthy adults,were measured to analyze the difference between each groups.we mainly explore the role of four factors,biological functions and in the occurrence and development of RA and their clinical significance.MethodThe samples of peripheral blood mononuclear cells used in this study were acquired from 30 patients and paired 30 healthy control.All of the 30 patients were diagnosed with RA and underwent internal medical treatment at Liaocheng Hospital between March 2018 and November 2018.To carry out this experiment,we firstly need to collect the general data of each group,ask the course of rheumatoid arthritis patients.Secondly,5 m L of elbow venous blood of each group was extracted with EDTA anticoagulation tube,and the cells were separated by human whole blood mononuclear cell separation solution;Automatic RNA purification instrument extracts total RNA;The concentration of RNA was detected by spectrophotometer and the integrity of RNA was detected by agarose gel electrophoresis;Reverse transcription of the RNA into c DNA by a gradient PCR machine;Design primers and complete real-time PCR.The data obtained were all recorded in the computer,using SPSS19.0 statistical sofware for statistical analysis.Result(1)Compared with the healthy group,the content of Hotair in peripheral blood mono-nuclear cells was significantly increased in the RA group,The differences between group were statistically significant(p <0.01).(2)Compared with the healthy group,the lncRNA-p21 content in peripheral blood mononuclear cells decreased significantly in the RA group,The differences between group were statistically significant(p <0.01).(3)Compared with the healthy group,the HCP5 content in peripheral blood mononuc-lear cells decreased in the RA group,The differences between group werenot statistically significant(p >0.05).(4)Compared with the healthy group,the content of RP1-68D18.2 in peripheral blood mononuclear cells increased in RA group,The differences between group werenot statistically significant(p >0.05).Conclusion(1)The level of Hotair in mononuclear cells of RA patients was significantly increased,and the level of lncRNA-p21 was significantly decreased.The difference was statistically significant,It is suggested that Hotair and lncRNA-p21 are involved in the regulation of the expression of encoded proteins,which may cause partial dysfunction of PBMC.These may be related to the onset and disease activity of RA.Further research may be a potential diagnostic marker for RA and an early therapeutic target.(2)The level of RP1-68D18.2 in mononuclear cells of RA patients increased,and the level of HCP5 showed an decreasing trend,but the difference was not statistically significant.Appropriate interventions may play an auxiliary role in the diagnosis and treatment of RA.
