| Objective: 5.5Gy ionizing radiation was used to establish an acute radiation brain injury model in adult Balb/c mice.Detect aerobic exercise intervention,aerobic exercise with Chinese traditional medicine epimedium intervention treatment of radiation brain injury such as cognitive function in mice and hippocampal dentate gyrus granular zone(subgranular zone,SGZ)proliferation and mature neurons(ki67,PV),and to explore different intervention measures to improve the effect of radiation brain injury,It provides some reference basis for the treatment of radiation brain injury.Method: The Animal Center purchased 40 6-week-old SPF Balb/c male mice and randomly divided them into a blank control group(N),a radiation control group(R-N),a radiation exercise traditional Chinese medicine group(R-E-epi),and a radiation exercise group(R-E),10 per group.After 2 weeks of adaptive feeding,except for group N,the other 3 groups of mice received acute X-ray radiation at a dose of 5.5 Gy.Within 2 hours after radiation,each group started to undergo aerobic exercise for 4weeks and intragastric administration of the traditional Chinese medicine Epimedium Wait for the corresponding treatment measures,and record the weight of the mice before irradiation as a reference baseline and weigh them at the same time every day thereafter.After 4 weeks of intervention,open field,tail suspension,Morris water maze and immunohistochemical experiments were performed to determine the weight gain,exercise ability,emotional state,learning and spatial memory and other cognitive functions of mice in each group,as well as ki67 and PV markers Related indicators such as the number of positive neurons.Results:(1)Body weight test: In the 5 weeks after radiation,the average increase in weight of mice was on the overall trend.The mice in the N group have been increasing,the RN group mice are in a negative growth trend,and the R-E-epi and R-E groups The mice showed a negative growth first and then a growth trend.In the 5weeks after radiation,compared with the N group,the average weight gain percentage of mice in the R-N and R-E groups was significantly lower than that in the N group(P<0.01),and the weight gain percentage of the mice in the R-E-epi group was significant in the first 2 weeks It was lower than the N group(P<0.05),and there was no significant difference in the next 3 weeks(p>0.05);compared with the R-N group,the average weight gain of mice in the R-E group was significantly higher than that in the R-N group in the 3-5 weeks(p <0.05),and there was no significant difference in the first 1-2 weeks(p>0.05).The average weight gain percentage of mice in the R-E-epi group was significantly higher than that in the R-N group in 5 weeks(P<0.05);compared with R-E The average weight gain of mice in the epi group and the R-E group was significantly lower than that in the R-E-epi group in the 4-5 weeks(p<0.05),but there was no significant difference in the 1-3 weeks(p>0.05).(2)Open field experiment: Compared with the R-N group,N,R-E-epi and R-E mice were significantly higher than the R-N group in the total distance,the percentage of the central movement distance,the central duration,the average movement speed,and the number of standings.(P<0.05),and there was no significant difference among the other groups(p>0.05).(3)Suspended tail test: Compared with the R-N group,the mice in the N,R-E-epi and R-E groups were significantly lower in resting time than the R-N group(p<0.05),and significantly higher in struggling times and climbing times than the R-N group(p<0.05).There was no significant difference among the other groups(p>0.05).(4)Morris water maze test: overall trend,as the experiment progressed,the average escape latency of mice in all groups except for the R-N group was generally shortened and relatively stable on the 3rd to 5th day.On the first day,the average escape latency of mice in each group was not significantly different(p>0.05).The average escape latencies of mice in the R-N group were significantly higher than those of the N group in the 2–5 days compared with those of the N group(p<0.01),the average escape latencies of mice in the R-E-epi group were significantly higher than those of the N group on the second day(p<0.05),but no significant difference was noticed on the 3–5days(p>0.05).The average escape latencies of mice in the R-E group were significantly higher than those of the N group on the 2–4 days(P < 0.05),but no significant difference was noticed on the fifth day(P > 0.05).Compared with the R-N group,the average escape latencies of mice in the R-E-epi group were significantly lower on the 3rd to 5th day than those in the R-N group(p<0.05)and there was no significant difference on the 2nd day(p>0.05).The average escape latencies of mice in the R-E group were significantly lower than those in the N group on the 5th day(p<0.05)and there was no significant difference on the 2nd to 4th day(P > 0.05).Compared with the R-E-epi group,the average escape latencies of the R-E group were significantly lower on the 3rd to 5th day than that of the R-E-epi group(p<0.05),and there was no significant difference on the 2nd day(p>0.05).In the space exploration experiment,compared with the R-N group,in the average number of times the mice cross the platform,R-E-epi and R-E>R-N are both statistically significant(P<0.05);compared with the R-E-epi group,in the mice On the average number of crossing the platform,R-E<R-E-epi and statistically significant(P<0.05).(5)mmunohistochemistry:Compared with group N,the expression numbers of ki67 and PV positive cells in SGZ region of mice in R-N group were significantly lower than group N(P < 0.01).Compared with the R-N group,the numbers of ki67 and PV positive cells in the R-E-epi and R-E groups were significantly higher than that in the R-N group(P < 0.01).Compared with the R-E-epi group,the expression numbers of ki67 and PV positive cells in the R-E group were significantly lower than those in the R-E-epi group(p<0.05).Conclusion:(1)5.5Gy ionizing radiation caused weight loss,emotional instability,and even depression in mice,significantly reduced exercise capacity and desire to explore,cognitive dysfunction such as learning and spatial memory,and proliferation of cells during neurogenesis in mice,Mature interneurons are significantly reduced.(2)4-week aerobic exercise,aerobic exercise and traditional Chinese medicine Epimedium intervention can all improve the weight loss,depression,reduced exercise ability,learning and memory related cognitive dysfunction in mice caused by ionizing radiation to a certain extent It can also protect the loss of newborn and mature neurons in mice SGZ and promote neurogenesis.(3)Combination therapy of aerobic exercise and traditional Chinese medicine epimedium is better than single aerobic exercise in improving nerve and
