Objective To investigate the expression and clinical significance of TIGIT on the membrane of T cells in patients with chronic hepatitis B.Methods Twenty-six patients with chronic hepatitis B(CHB)who received antiviral treatment for the first time were collected as the study group,and Twenty healthy person were taken as the control group.The study group was given antiviral treatment with entecavir tablets.According to the treatment time,they were divided into CHB antiviral pre-group,CHB antiviral 4 weeks group,and CHB antiviral 12 weeks group.Peripheral venous blood of each group of study subjects was collected.Peripheral blood mononuclear cells(PBMC)and plasma were separated by density gradient centrifugation and low-speed centrifugation.Flow cytometry was used to detect the expression of T cell immunoglobulin and immunoreceptor tyrosine inhibitory motif domain(TIGIT)on the membranes of CD4~+T cells and CD8~+T cells in PBMC,and ELISA was used to detect plasma cytokines(IL-10,IFN-γand TNF-α)levels,and at the same time,detect the ALT,HBV DNA quantification and hepatitis B three systems in the liver function of CHB patients.IBM SPSS 26.0 software was used to analyze the data to analyze the results of the study statistically,and P<0.05 was statistically significant.Results1.The percentages of CD4~+T and CD8~+T cells in the peripheral blood of patients in the CHB antiviral 0 weeks group were significantly lower than those in the control group(P<0.05).The expression of TIGIT on the membranes of CD4~+T cells and CD8~+T cells in CHB patients was significantly increased.Compared with the control group,the difference was statistically significant(P<0.05).The expression level of TNF-αin plasma was increased,and the expression of IFN-γand IL-10 was low.Compared with the control group,the difference was statistically significant(P<0.05).2.The research group was given antiviral therapy.With the extension of antiviral time,the decrease of HBV DNA load and the improvement of liver function,the expression of TIGIT on the membranes of CD4~+T cells and CD8~+T cells gradually decreased,and the difference between the groups was statistically significant(P<0.05).The levels of IFN-γand IL-10 in the patients’plasma gradually increased,and the level of TNF-αgradually decreased,and the difference between the groups was statistically significant(P<0.05).The membrane changes of TIGIT~+CD4~+T cells and TIGIT~+CD8~+T cells are positively correlated with the changes of HBV DNA,ALT and TNF-α,negatively correlated with IFN-γ(all P<0.05),and have no change with IL-10 Correlation(P>0.05).Conclusion1.The high expression of inhibitory receptor TIGIT on the peripheral blood T cell membrane of patients with chronic hepatitis B.2.The high expression of inhibitory receptor TIGIT on the peripheral blood T cell membrane of patients with chronic hepatitis B has a certain correlation with plasma viral load and inflammatory indicators. |
