Objective:To study the changing regularity of the pathology and the expression of P-p38 MAPK,p62 in the diaphragm of acute chlorpyrifos poisoning rats,in order to provide a theoretical basis for further elucidating the pathogenesis of intermediate syndrome.Methods:In this experiment,the rats were administered with81.5mg/kg(1/2LD50)chlorpyrifos by gavage to establish acute chlorpyrifos poisoning rat model.35 male SD rats were randomly divided into normal control group(Control),1/2 day group,1 day group,2 day group,3 day group,5day group,and 7 day group(1/2d,1d,2d,3d,5d and 7d),with 5 rats in each group.Each poisoning group rats were poisoned by chlorpyrifos-olive oil,sacrificed and removed the diaphragm after 1/2d,1d,2d,3d,5d and 7d.The normal control group rats were sacrificed and removed the diaphragm 3 days later after gavage with olive oil.Observing the symptoms of acute chlorpyrifos poisoning rats.Using HE-staining to observe the histopathological changes of diaphragm.Detecting the expression level of P-p38 MAPK and p62 in the diaphragm tissues of acute chlorpyrifos poisoning rats by immunoblotting.Results:Compared with the normal control group,the rats of each poisoning group showed significant acute organophosphorus pesticide poisoning symptoms and some degree of respiratory dysfunction,reaching a peak in 12-24 hours after the poisoning.HE staining showed that acute chlorpyrifos poisoning caused structural changes in the diaphragm muscles of rats,reached the most severe injury in the third day.Immunoblotting showed that:with the passage of time,the expression of P-p38 MAPK protein in the diaphragm of rats with acute chlorpyrifos poisoning showed a sharp increase and then decreased,reaching a peak in 1/2 day,the difference was statistically significant(P<0.05).While the expression of p62 protein showed a gradual increase and then decreased,reaching a peak in 3 days,the difference was statistically significant(P<0.01).There was no significant change in the expression of p38 MAPK protein(P>0.05).Conclusion:In the early stage,acute chlorpyrifos poisoning activated the p38 MAPK signaling pathway and induced the accumulation of p62 protein in the rats diaphragm.The high expression stage of p62 and the most severe stage of diaphragm pathological damage were consistent with the onset time of intermediate syndrome.we speculated that the accumulation of p62 and the diaphragm injury might be related to intermediate syndrome.
