| Objective: Establish a rat liver regeneration model to explore the effect of dual arterial blood supply on oxidative stress in liver regeneration after partial hepatectomy.Methods: 1.108 clean-grade male SD rats were randomly divided into three groups: 68%hepatectomy(partial hepatectomy,PH group),control group(Control group,CG group),68%hepatectomy combined with dual hepatic arterial blood supply(liver dual arterial blood supply,LDABS)group(LDABS group for short).The animal models of each group were taken at the time points 0,12,24,72,120 and 168 h after the operation.Measure malondialdehyde(MDA)content,superoxide dismutase(SOD)content,glutathione peroxidase(GSH-Px)content,and Western blot to detect PINK1(PTEN induced putative kinase 1),Bcl-2(B-cell lymphoma 2)Protein expression.2.SPSS22.0 analysis and(?)±s were used to describe the measurement data.One-way ANOVA was used to analyze the variance.LSD test was used to compare the mean between the two groups.When the variance was uneven,the rank sum Mann-Whitney U test was used to compare the two rates.Chi-square test was used to compare the two rates,P < 0.05.it was considered that there was significant difference.Results:(1)Compared with the CG group,the MDA content in the liver tissue of rats in the PH group increased from 0.5 to 3 days after treatment,and the MDA content in the liver tissue of rats in the LDABS group increased from 0.5 to 5 days after treatment,but there was no significant increase Difference: Compared with the PH group,the MDA content of the LDABS group increased from 0-7 days,but there was no significant difference.(2)Compared with the CG group,the SOD content in the liver tissue of rats in the PH group decreased at 0.5-7 days after treatment,and the SOD content in the liver tissue of rats in the LDABS group decreased at 0.5-7 days after treatment.There was a significant difference on the 7th day(P<0.05);compared with the PH group,the SOD content of the LDABS group increased in 3-5 days,but there was no significant difference.(3)Compared with CG,the GSH-Px content in the liver tissue of the PH group decreased from 1-7 days after treatment,and there was a significant difference between 3-5 days,and the liver tissue of the LDABS group was 3-7 days after treatment.The GSH-Px content decreased;compared with the PH group,the GSH-Px content of the LDABS group increased on1 and 5 days,but there was no significant difference.(4)Compared with the CG group,the Bcl-2protein expression level in the LDABS group was significantly increased(P<0.05);the PINK1 protein expression in the PH group was increased compared with the CG group,and the PINK1 protein expression in the LDABS group was slightly higher.Decrease,but there is no significant difference.Conclusions: LDABS did not adversely affect the oxidative stress of liver regeneration after partial hepatectomy compared with PH,and it can promote the expression of Bcl-2 protein compared with CG group. |
