Objective In this study,immunohistochemical(Immunohistochemical,IHC)and Western-Blot(Western-Blot,WB)were used to test the preoptic area of the hypothalamus(POAH)of experimental rats by estrogen and adrenergic receptors.The relationship between body expression was tested to explore the central neurotransmitter mechanism of hot flashes in perimenopausal women and the possible mechanism of action and treatment for the clinical use of estrogen replacement therapy(ERT)to relieve hot flashes The role provides a more adequate theoretical basis,and then provides new evidence for the clinical use of ERT therapy to treat perimenopausal hot flashes.Methods 30 female SD(Sprague-Dawley,clean-grade rats)rats were randomly divided into OVX(ovary removed group),SHAM(sham operation group without ovarian removal),OVX+E2(estrogen supplementation treatment group after ovarian removal))Three groups.The success of the model was confirmed by taking vaginal exfoliated cell smears,measuring estrogen levels,measuring uterine wet weight,and measuring the temperature of the rat’s tail.Two methods,IHC and WB,were used to detect the expression of adrenergic α1 and α2 receptors in the preoptic area of the hypothalamus and the levels of estrogen receptor(Estrogen receptor,ER)α,β and GPR30(G Protein-coupled receptor 30,ER)What is the relationship between the amount of expression.All statistical processes are completed using statistical software SPSS21.0.Results The experimental model was successful.1.The number of adrenergic α1receptor cells in the hypothalamic preoptic area of the OVX group was less than that of the SHAM group(p﹤0.05),and the number of adrenaline α2 receptor cells was more than that of the SHAM group.2.The number of adrenaline α1 receptor cells in the OVX+E2 group was higher than that in the OVX group,and the number of adrenalineα2 receptor cells was less than that in the OVX group(p﹤0.05).3.The expression of ERα and β receptors in the preoptic area of the hypothalamus in the OVX group was significantly lower than that in the SHAM group(p﹤0.05),and the expression of ERα and β receptors in the preoptic area of the hypothalamus in the OVX+E2 group Significantly more than the OVX group(p﹤0.05).And the results of the above groups were not statistically significant compared with the SHAM group(p﹤0.05).4.There was no statistically significant difference in GPR30 expression between the three groups of rats(p﹥0.05).Conclusion The experimental model was successful.1.The number of adrenergicα1 receptor cells in the hypothalamic preoptic area of the OVX group was less than that of the SHAM group(p﹤0.05),and the number of adrenaline α2 receptor cells was more than that of the SHAM group.2.The number of adrenaline α1 receptor cells in the OVX+E2 group was higher than that in the OVX group,and the number of adrenalineα2 receptor cells was less than that in the OVX group(p﹤0.05).3.The expression of ERα and β receptors in the preoptic area of the hypothalamus in the OVX group was significantly lower than that in the SHAM group(p﹤0.05),and the expression of ERαand β receptors in the preoptic area of the hypothalamus in the OVX+E2 group Significantly more than the OVX group(p﹤0.05).And the results of the above groups were not statistically significant compared with the SHAM group(p﹤0.05).4.There was no statistically significant difference in GPR30 expression between the three groups of rats(p﹥0.05). |
