| Objective To clarify the interaction between NDRG2 and NF-κB-mediated apoptosis signaling pathway in acute intracranial hypertension after rabbits with severe craniocerebral injury were treated with controlled decompression technique by animal experiments,to provide the basis for further exploring the mechanism of controlling decompression.Methods 60 healthy adult New Zealand white rabbits were randomly divided into controlled decompression group(n=20),fast decompression group(n=20)and sham operation group(n=20).At 24 hours after operation,the behavioral score of each group was evaluated,the water content of brain tissue,the expression levels of NDRG2,NF-k B and Caspase-3 molecules were detected,and the apoptosis of nerve cells at the operation site was detected by TUNEL fluorescence staining.Results Compared with the sham group,the behavioral scores in the controlled decompression(4.50 ± 0.60)group and the fast decompression group(7.10 ± 0.52)were higher than those in the control group(3.30 ± 0.34)(p < 0.05).The water content in brain tissue after operation,NF-κB and Caspase-3 expression levels increased,but the expression level of NDRG2 decreased.TUNEL fluorescence staining showed that the degree of neuronal apoptosis increased significantly.Compared with the fast decompression group,the animal behavior score in the controlled decompression group was lower than that in the fast decompression group(p < 0.05).The water content in brain tissue after operation,NF-κB and Caspase-3 expression levels were lower than those in the fast decompression group,but the expression level of NDRG2 was higher.TUNEL fluorescence staining showed that the degree of neuronal apoptosis was significantly reduced.Conclusion Compared with traditional operation,controlled decompression operation can improve the prognosis of the disease.Step-wise decompression is used to gradually increase cerebral perfusion pressure,thus reducing the occurrence of cerebral ischemia-reperfusion injury in secondary brain injury,decreasing the degree of neuronal apoptosis and relieving the degree of cerebral edema after craniocerebral injury.In this experiment,the expression of NDRG2 in the controlled decompression group was higher than that in the fast decompression group.However,the expression of NF-κB and Caspase-3 in the fast decompression group was decreased compared with that in the fast decompression group,and there was a negative correlation.Meanwhile,it is further verified that NDRG2 plays an important protective role in nervous system diseases,which may result from the fact that NDRG2 expression is increased by controlled decompression surgery to inhibit the generation of NF-κB mediated apoptosis signaling pathway,thus playing a protective role on nerve cells after severe craniocerebral injury. |
