BackgroundPolycystic ovary syndrome(PCOS)has complicated causes and unclear pathogenesis.Studies have shown that the abnormal expression of rapamycin target protein(mammalian target of rapamycin,mTOR)may be related to the pathogenesis of PCOS[1].mTOR is a serine/threonine protein kinase that is constantly expressed in cells[2].Once it is activated,phosphorylated mTOR(P-mTOR)becomes active[3].At present,most of the research on the relationship between mTOR and PCOS is limited to in vitro ovarian cell culture or animal experiments,and there are few reports on its research in PCOS patients.Therefore,this study aims to explore the expression of serum mTOR in patients with PCOS,in order to provide theoretical support for the realization of PCOS targeted therapy.ObjectiveBy detecting the concentration of P-mTOR and mTOR in the serum of PCOS patients,the correlation between P-mTOR and mTOR and obesity,sex hormone levels,and glucose metabolism was analyzed.Provide potential targets for the follow-up in-depth study of the pathogenesis and treatment of PCOS.MethodsWe selected 80 patients with PCOS who were diagnosed with PCOS in the Department of Obstetrics and Gynecology,the First Affiliated Hospital of Xinxiang Medical College between January 2019 and July 2020.During the same period,80 women with normal ovarian function reserve were used as the control group.Collect general information of the research subjects,such as hip circumference,waist circumference,height,weight,age,systolic blood pressure,diastolic blood pressure,etc.use enzyme-linked immunosorbent assay to detect serum p-mTOR and mTOR protein concentrations.use electrochemiluminescence immunoassay Method to detect serum luteinizing hormone(LH),follicle stimulating hormone(FSH),testosterone(T),estradiol(E2),progesterone(P),prolactin(PRL),fasting insulin(FINS)levels;application Glucose oxidase method to detect fasting blood glucose(FPG).calculate the patient’s body mass index(BMI),waist-to-hip ratio,LH/FSH,insulin resistance index(HOMA-IR),and compare all test results between groups.Results1 BMI,P-mTOR,LH,T,LH/FSH,and HOMA-IR in the PCOS group were significantly higher than those in the control group,and the difference was statistically significant(P<0.05);PCOS group age,hip circumference,waist circumference Compared with the control group,there was no significant difference in waist-to-hip ratio,systolic blood pressure,diastolic blood pressure,mTOR,FSH,E2,P,PRL,and FPG(P>0.05);2 The p-mTOR level of PCOS patients was positively correlated with T and HOMA-IR(P<0.05),but not correlated with BMI(P>0.05);3 There is no correlation between mTOR level in PCOS patients and T,HOMA-IR and BMI(P>0.05).Conclusions1 There is abnormal phosphorylation of mTOR in patients with PCOS;2 The pathogenesis of PCOS may be related to the functional status of the mTOR protein,and has nothing to do with the level of expression;3 Abnormal phosphorylation of mTOR in patients with PCOS is significantly positively correlated with changes in T and IR.It is speculated that the three may be involved in the occurrence and development of PCOS. |
