| Objective:We intend to study the relative expression levels of PRMT5 and PRMT7 in renal cancer tissues and normal tissues adjacent to the cancer,and the relationship between their overexpression in renal cancer and clinical indicators,pathological indicators,and survival prognosis of renal cancer.In-depth discussion of the feasibility of PRMT5 and PRMT7 as a predictor of the progression of renal cancer and as a predictor of the prognosis of renal cancer.Methods:After passing the application and obtaining the approval of the hospital ethics review committee and obtaining the informed consent of all patients and their families,we collected 51 cases of renal cell carcinoma diagnosed in the Second Affiliated Hospital of Nanchang University,Jiangxi Province from 2015 to 2020,and postoperative pathological reports of all patients Confirmed to be renal cell carcinoma.All selected patients completed follow-up and obtained complete data.Among them,there were 26 male patients and 25 female patients,aged between 26 and 76 years old.All the pathological specimens of the selected patients were resectioned and then subjected to immunohistochemical examination to observe the expression differences of PRMT5 and PRMT7 in renal cell carcinoma,and parallel semi-quantitative analysis.Finally,the data of the patients were statistically built and the database was established.The relationship between the expression levels of PRMT5 and PRMT7 and various clinical pathological indicators was analyzed by the 2 test.The survival rate was analyzed by the Kaplan-Meier method,and the log was used for the univariate survival analysis.-rank test,using Cox multivariate model analysis method to screen the factors that have a significant impact on survival time,using R language rms package to draw a prognostic prediction nomogram,P<0.05 is considered statistically significant,and R4.0.2 is used for all Statistical Analysis.Results:Among all 51 patients in this trial,there were 26 male patients(50.9%)and 25 female patients(49.1),with an average age of 53.26 years,the youngest age was 26 years old,and the oldest was 77 years old.There are 6 cases(7.8%)of TNM stage 4cases,5 cases(9.8%)with lymph node metastasis,and 4 cases(7.8%)with distant metastasis.The 1-year,3-year,and 5-year survival rates of 51 patients were 96.1%,86.3%,and 74.5%,respectively.The results of immunohistochemical examination of renal cancer tissues and normal tissues adjacent to the cancer found that the negative and positive expression rates of PRMT5 renal cancer tissue were 9.8% and 90.2%,respectively,and positive expressions were +(29.4%),++(45.1%),+ ++(15.7%),the negative and positive expression rates of PRMT7 renal cancer tissue were 5.9% and94.1%,respectively,and positive expression was +(21.6%),++(54.9%),+++(17.6%).PRMT7 is mainly stained in the cytoplasm and nucleus of tumor cells.In 51 cases of normal kidney tissues adjacent to cancer,PRMT5 and PRMT7 are mostly negatively expressed,and a few weakly positive expression rates are 17.6%(PRMT5),23.5 %(PRMT7),no case of strong positive expression,the difference in expression levels between renal cell carcinoma tissue and normal tissue adjacent to the cancer was statistically significant(P<0.01).This study found that the patient’s gender,age,tumor side,and tumor Fuhrman grade There is no correlation with the expression levels of PRMT5 and PRMT7(P>0.05),while the four indicators of tumor size,TNM stage,lymph node metastasis,and distant metastasis are positively correlated with the expression levels of PRMT5 and PRMT7,and the difference is statistically significant(P<0.01).Multivariate analysis of survival analysis showed that PRMT5 expression intensity,PRMT7 expression intensity,lymph node metastasis,distant metastasis,TNM staging,Fuhrman grade,tumor maximum diameter and other factors were related to the prognosis of renal cancer,and the difference was statistically significant.Further COX regression analysis found that the maximum tumor diameter ≥ 7cm(3.238),TNM staging(HR4.148),lymph node metastasis(HR5.238),distant metastasis(HR9.204),PRMT7 expression level(HR3.774),etc.Factors are independent risk factors for the prognosis of renal cancer,among which distant metastasis particularly affects the prognosis of renal cancer.Finally,we drew a nomogram to predict the prognosis of kidney cancer.Conclusion:1.The expression levels of PRMT5 and PRMT7 are positively correlated with the maximum tumor diameter,TNM stage,lymph node metastasis,and distant metastasis.2.PRMT5 expression level,PRMT7 expression level,lymph node metastasis,distant metastasis,TNM stage,Fuhrman grade,tumor maximum diameter and other factors are negatively correlated with the prognosis of renal cell carcinoma.Among them,PRMT7 expression level,lymph node metastasis,distant metastasis,tumor maximum diameter ≥7cm,TNM staging and other five factors are independent risk factors for the prognosis of renal cancer,and the risk ratio of distant metastasis is the highest.3.At the end of this study,a nomogram for predicting the prognosis of kidney cancer is provided. |
