Objective:To explore the effect of exogenous sodium hydrosulfide on the function of endothelial cell contraction and relaxation in burned rats and the related mechanism.Methods: The neonatal rat abdominal aorta endothelial cells were prepared in vitro and cultured to the third passage for use.Select 350-400 g of the abdominal aortic blood of healthy male SD rats and burned SD rats after centrifugation and mark them respectively,and filter them with a 300-mesh filter to prepare normal serum and burned rat serum for use.The experimental groups are normal respectively.Serum,burn serum and burn serum + NaHS(Sodium hydrosulfide,NaHS),a total of 3 groups.There are 4 concentrations of NaHS: 50,100,150,200μmol/L.The third-generation endothelial cells of neonatal rats cultured in vitro,after synchronization treatment,were grouped and intervened for 4 hours according to the above experiment,and the cell supernatant was collected.Enzyme linked immunosorbent assay(ELisa)and nitrite quantification method(Griess)were used to determine the corresponding reactants of endothelial function of nitric oxide(NO)and endothelin-1(Endothelin-1).,ET-1)expression level.Results:Intervene in different groups at the same time node,and detect the expression levels of NO and ET-1 in rat endothelial cells in each group.It can be concluded that:(1)Compared with normal serum intervention endothelial cell group,burn serum group and burn serum + NaHS intervention,the expression levels of NO and ET-1 in the endothelial cell group were significantly increased,and the difference was statistically significant(p<0.05);(2)The expression of NO in the burn serum +NaHS intervention endothelial cell group was significantly higher than that in the burn serum intervention group.It is statistically significant(p<0.05);(3)The concentration of ET-1 in the burn serum + NaHS intervention endothelial cell group was significantly lower than that in the burn serum intervention endothelial cell group,and the difference was statistically significant(p<0.05));(4)50~200umol/l NaHS promotes the secretion of NO by rat endothelial cells,but NaHS at the same concentration always inhibits the production of ET-1;Conclusions:After burn stress,endothelial cell diastolic and contractile function can be affected by trace exogenous hydrogen sulfide.This change in diastolic function is caused by the decreased expression of ET-1 and increased expression of NO caused by trace exogenous NaHS.
