Objective:To explore the relationship between the methylation levels of COL4A1,ANGPT1 proximal promoter region and 5 ’-UTR region and Budd Chiari syndrome,and to provide a theoretical basis for the etiology,diagnosis and treatment of Budd Chiari syndrome.Method:Six patients with Budd Chiari syndrome admitted to the Department of vascular surgery of the First Affiliated Hospital of Bengbu Medical College from October 2018 to A pril 2019 were selected as the experimental group;six healthy subjects with no significa nt difference in age distribution and gender ratio in the physical examination center during the same period were selected as the control group.Venous blood samples were obtained,DNA was extracted,sample processing,primer design and application of Agen a massarray Methylation were performed Detection and mass spectrometry analysis,through the mass spectrum to determine the difference in molecular weight,methylation analysis.Result1.A total of 149 Cp G loci were detected by 8 primers of COL4A1 and ANGPT1 genes.2.There were differences in the methylation levels of Cp G_6 and Cp G_20 in the primers COL4A1_20 in the methylation map,and there was no statistically significant difference in the methylation levels(P > 0.05).In primer COL4A1_22,there were differences in the degree of methylation between experimental group and control group on Cp G_7 and Cp G_13.The difference of methylation level was statistically significant(P≤0.05).There was no significant difference in methylation level at other primer sites between the two groups.3.In primer COL4A1_22,the levels of Cp G_7 and Cp G_13 in the experimental group were lower than those in the control group.ConclusionHypomethylation of some fragments in the COL4A1 promoter region and5’-UTR region may cause high gene expression;suggesting that hypomethylation of some fragments in the COL4A1 promoter region and 5’-UTR region may be one of the pathogenic factors of BCS in northern Anhui.
