| Objective:To study the expression of focal adhesin kinase(FAK)in the eyelid Müller muscle of patients with thyroid-associated ophthalmopathy(TAO),and to provide an effective research target for the pathogenesis of thyroid-associated ophthalmopathy eyelid retraction.Methods:From December 2017 to December 2019,8 patients with thyroid-related ophthalmopathy and 8 patients with ptosis were diagnosed and undergone surgical treatment at the Tianjin Medical University Eye Hospital.1.Masson staining was used to observe the degree of Müller muscle fibrosis.2.Immunohistochemical staining of all specimens,including α-SMA antibody,FAK antibody,p-FAK antibody,β-catenin antibody,and vinculin antibody staining,to detect the protein expression in Müller muscle.3.Using Western-blot to detect the expression of FAK,p-FAK,β-catenin and vinculin protein.4.Extract the total RNA from the muscle specimens obtained by surgery,and apply Real-time Quantitative polymerase chain reaction(RT-q PCR)after reverse transcription to detect TGF-β1,FAK,β-catenin,TGF in Müller muscle-Gene expression of TGF-β1,TGF-β2,TGF-βR1,TGF-βR2.Results:1.Masson staining results of specimens showed that the control group had normal Müller muscle structure,and the patients with thyroid-related ophthalmopathy had obvious fibrosis.2.Compared with the control group,the Müller muscle of TAO patients showed fibrotic changes.The α-SMA in the muscle tissue suggested that the tissue was smooth muscle.FAK,p-FAK and vinculin were significantly overexpressed,and the antibody staining degree of β-catenin in the two groups No significant differences were seen.3.Western-blot test results showed that FAK,p-FAK and vinculin were highly expressed in the TAO group,while β-catenin protein expression was not different between the two groups.4.RT-q PCR results showed that the expressions of TGF-β1 m RNA、 FAK m RNA and vinculin m RNA in the eyelid Müller muscles of patients with thyroid-related ophthalmopathy were significantly higher than those in the control group,and the expression levels of the two were correlated.TGF-βR1 and TGF-βR2 m RNA were also Significantly increased,TGF-β2 m RNA expression was not different from the normal group,and β-catenin m RNA expression in the two groups was also no difference.Conclusion:1.Müller muscle interstitial fibrosis changes in the eyelid of patients with thyroid-related ophthalmopathy.2.Its formation mechanism is related to TGF-β1 / FAK. |
