Objective:In this experiment,a mouse model of osteoporosis was established and TSA was used for treatment throughout the body to study the effect of TSA on bone metabolism under osteoporosis.It is hoped that this will be a foothold for the prevention and treatment of oral diseases in osteoporosis patients Provide new ideas and treatment targets.Methods:Thirty balb/c mice aged 6 weeks were selected and divided into sham operation group(sham group,n=10)and osteoporosis model group(OVX group,n=20)by random number table method.In the OVX group,the ovary was completely removed.5% chloral hydrate is used to anesthetize mice at 6ml/kg,and the method of anesthesia adopts the conventional intraperitoneal drug injection method.The anesthesia effect is accurate for the back skin.A longitudinal incision of 0.5-1 cm is made in the middle of the dorsal part of the lower abdomen.Use sterile forceps to bluntly separate the two sides of the spine along the middle of the back.Pull out the ovarian ligation and remove it.The incision was sutured layer by layer and penicillin was given to prevent infection after surgery.In the sham group,only fat was removed,and the size was equivalent to the ovarian volume.Weighed every week,and after 8weeks of modeling,tissue slices and Micro CT were used to detect the modeling effect.After successful modeling,10 mice of the OVX group were randomly selected for intraperitoneal injection of TSA(TSA group,n=10,the dosage was 5mg/kg,once every other day,a total of 4 times)one month after the medication(serum frozen;visceral fixation;bone tissue including alveolar bone,humerus,femur,tibia,frozen on one side,fixed on one side).Micro CT,HE conventional staining,RT-PCR and other F were used to study the effect of TSA on bone metabolism and the expression of bone metabolism related molecules.Result:After removing the ovaries,the bone strength of the mice is reduced,and the structure becomes weaker.The rate of new bone formation is reduced and the formation is reduced.TSA can increase the new bone formation ability of ovariectomized mice by up-regulating the key genes of bone formation Runx2,Sp7,Opg,and down-regulating the key genes of osteoclast Rankl.Has obvious therapeutic effect.Conclusion:In ovariectomized mice,the bone microstructure was destroyed,the rate of new bone formation decreased,and bone formation decreased.TSA can increase the new bone formation of ovariectomized mice,improve bone microstructure,and have a therapeutic effect on osteoporosis by up-regulating Runx2,Sp7,and Opg,the key genes for osteogenesis in bone tissue,and down-regulating the osteoclast gene Rankl in OVX mice. |
