| China is the most populous country in the world.Due to the improvement of quality of life,non-alcoholic fatty liver disease(NAFLD)is becoming more and more common.The pathogenesis of NAFLD has not yet been fully clarified.Object:Through clinical examination and anthropometric evaluation,NAFLD is closely related to body fat,obesity,hyperlipidemia and impaired fasting blood glucose.Methods:Through clinical examination and anthropometric evaluation,NAFLD is closely related to body fat,obesity,hyperlipidemia and impaired fasting blood glucose.A Kunming(KM)mouse model of NAFLD was established.90 KM mice were randomly divided into 9 control groups and 9 high-fat groups,and mice were obtained on days 7,10,15,18,21,24,28,31 and 35,respectively serum was used for clinical biochemical determination,and liver slices were taken for histological evaluation.GC-MS and HPLCMS targeted metabolomics methods were used to analyze serum lipid metabolism in the population of NAFLD and mice,followed by unsaturation analysis.NMR non-targeted metabolomics analysis of NAFLD mouse plasma,and then metabolic pathway analysis.Results:NAFLD was closely related to body fat,obesity,hyperlipidemia and impaired fasting blood glucose,and the content of OAHOAs changed significantly.The body weight of the high-fat diet(HFD)group was significantly higher than that of the control group.The levels of serum aminotransferase,glucose,total cholesterol,and triglycerides were significantly higher than that of the control group,and the levels of total protein and albumin were lower than that of the control group.The liver in the HFD group was dull,albino,and brittle in texture,and the liver volume was larger than that in the control group.At nine time points in the HFD group and the control group,the content of palmitoleic acid,oleic acid,docosahexaenoic acid,glycerin,glucose,betaine,lysine,glutamine,glutamic acid,valine,alanine,arginine,choline,and leucine has significant changes.Monounsaturated fatty acids(MUFA)and polyunsaturated fatty acids(PUFA)changes are obvious.The metabolism of alanine,aspartic acid and glutamic acid,the metabolism of D-glutamine and D-glutamic acid,and the biosynthetic pathway of arginine are most affected by significantly different metabolites.Based on the combination of clinical investigations,animal models,and metabolomics studies on the NAFLD population and the KM mouse model,OAHOAs,palmitoleic acid,oleic acid,docosahexaenoic acid,glycerin,glucose,betaine,lysine,glutamine,glutamate,valine,alanine,arginine,choline,leucine,MUFA,PUFA are biomarkers of NAFLD,by affecting alanine,aspartic acid and glutamate Metabolism,D-glutamine and D-glutamate metabolism,arginine biosynthesis pathway and lipid metabolism,affect the development of NAFLD. |
