| Objective: The expression level of mi RNA-182 in osteosarcoma HOS cells was detected,and the effect on the apoptosis of osteosarcoma HOS cells was determined by constructing an overexpression mi RNA-182 transfection vector.Methods : The expression level of mi R-182 in osteosarcoma HOS cells was detected by RT-q PCR.A transfection vector of over-expressed mi RNA-182 was constructed and the cells were co-cultured after transfection.The expression level of mi RNA-182 after transfection was detected by RT-q PCR,and the cell morphology was observed under a microscope.Western blot analysis was performed to explore the effect of mi R-182 on the RAC1 pathway protein.Cell Counting Kit-8(CCK-8)was used to analyze the effect of mi RNA-182 on the proliferation activity of osteosarcoma HOS cells.To evaluate the effects of mi R-182 on the cell cycle and apoptosis of osteosarcoma HOS cells.Results:Mi RNA-182 is underexpressed in osteosarcoma HOS cells.After the over-expressed mi RNA-182 transfection vector was constructed,the expression levels of RAC1 pathway protein and downstream PAK1 protein in osteosarcoma HOS cells were significantly reduced.Overexpression of mi RNA-182 significantly promotes the apoptosis of osteosarcoma cells and prevents their cell cycle progression.Conclusions: Mi RNA-182 is underexpressed in osteosarcoma.Regulation of the RAC1 pathway can promote osteosarcoma HOS cell apoptosis and prevent cell cycle progression. |
