| Objective: Construct a moisture balanced wound dressing with antibacterial properties to prevent bacterial infection on the wound and provide an appropriate microenvironment for wound healing,so as to promote wound healing.Propose a new plan for the development of wound dressings for clinical wound treatment.Methods: The dressing moisture balance test explored the moisture balance performance of the polyurethane foam dressing,and the moisture balanced wound healing experiment evaluated the optimal wound dressing humidity.We constructed a moisture balanced antibacterial dressing by loading lysozyme onto a polyurethane foam dressing,named PU/DA-LYS(polyurethane foam dressing/dopamine-lysozyme).The synthesis of PU/DA-LYS was evaluated by testing the physical and chemical properties of the dressing.The water contact angle experiment evaluated the hydrophilic properties of PU/DA-LYS.The drug loading test experiment evaluated the drug loading of PU/DA-LYS loaded lysozyme.The drug release curve evaluated the sustained release performance of PU/DA-LYS on lysozyme.The bacterial suspension method tested the in vitro antibacterial properties of PU/DA-LYS.The cytotoxicity test evaluated the biocompatibility of PU/DA-LYS.The infection wound healing experiment evaluated the antibacterial properties and wound healing effects of PU/DA-LYS in vivo.Anatomical image of animal viscera after infected wound healing experiment evaluated the in vivo biocompatibility of PU/DA-LYS.Results: The dressing moisture balance test showed that the polyurethane foam dressing showed a balance of absorption and loss of water on the wound surface,so it had the condition of being a moisture balanced wound dressing.Moisture balanced wound healing experiment shown that wound healing was the fastest when the polyurethane foam dressing humidity was 25%.The characterization results of the dressing shown that the synthesis of PU/DA-LYS was successful,and it has good hydrophilic properties and sustained drug release properties.In vivo and in vitro antibacterial experiments show that PU/DA-LYS has excellent antibacterial properties.Experiments on wound healing in infected mice shown that PU/DA-LYS can prevent wound infection and promote wound healing.Cytotoxicity experiments and in vivo biological safety experiments show that PU/DA-LYS was safe.Conclusion: We constructed a moisture balanced antibacterial dressing(PU/DA-LYS)that can not only prevent bacterial infection but also provide a suitable humidity environment for wound healing,with good biocompatibility to be a promising wound dressing. |
