| Objective:In this study,we investigate the level of serum silencing information regulators 6 in different glucose tolerance subjects,and explore the correlation between SIRT6 and glucolipid metabolism,obesity and insulin resistance.Methods:A total of 280 volunteers who received physical examination in the outpatient clinic of the endocrinology department of Hebei General hospital from January to December 2019 were selected as the research subjects.After signing the written informed consent form,all subjects underwent routine physical examination,including the measurement of height,weight,waist circumference,hip circumference,blood pressure and electrocardiogram;blood,urine and stool routine;meanwhile,their venous blood samples were taken to measure biochemical items,insulin,glycosylated hemoglobin(HbA1c)and other indicators.All participants received 75g oral glucose tolerance test(OGTT),their homeostasis model assessment ofβ-cell function(HOMA-β)and homeostasis model assessment of insulin resistance(HOMA-IR)were calculated simultaneously.According to the results of glucose tolerance test,the subjects were divided into three groups,including normal glucose tolerance(NGT),impaired glucose regulation(IGR)and type 2 diabetes mellitus.The SIRT6 level in participants with different glucose tolerance were determined by enzyme-linked immunosorbent assay(ELISA).SPSS21.0software was used for all statistical analysis.Results:The serum SIRT6 levels showed a decreasing trend among NGT,IGR and T2DM groups,and we found there was significant difference among these three groups.In the total population,compared with the non-obese group,the obese group had lower SIRT6 level(P<0.01).The serum SIRT6 concentration was the lowest in the obese subgroup of T2DM.When the subjects were divided into male and female groups,the serum level of SIRT6 in male was lower than that in female,and their concentrations were respectively(P<0.01).Correlation analysis showed that serum SIRT6 was positively associated with HOMA-β(P<0.01).The level of serum SIRT6 was negatively correlated with age,BMI,WHR,FPG,2hPG,HbA1c,HOMA-IR and FIns(P<0.05 or P<0.01).After further adjustment for gender,age,BMI and WHR,the level of serum SIRT6 was still positively correlated with HOMA-β(P<0.01),the serum SIRT6 was negatively correlated with FPG,2hPG,HbA1c and HOMA-IR(P<0.01).Multiple stepwise regression analysis indicated that 2hPG was an independent influencing factor for serum SIRT6 level(R~2=0.434,P<0.01),SIRT6,FPG,HOMA-β,FIns,2hIns and HDL-C were important influencing factors for HOMA-IR.SIRT6,FPG,HOMA-IR and FIns were important influencing factors for HOMA-β.Binary Logistic regression analysis indicated that even after adjusting for the basic parameters(age,gender,BMI,WHR)and the related factors such as lipid profile,low SIRT6level might be a risk factor for T2DM(P<0.01).Conclusion:1 The serum SIRT6 level decreased gradually with the aggravation of glucose tolerance impairment,which was the lowest in the T2DM group,especially in the subgroup of T2DM with obesity,the serum SIRT6 showed a negative correlation with blood glucose and HbA1c levels.2 The serum SIRT6 was negatively correlated with HOMA-IR and positively correlated with HOMA-β,which was an important influencing factor of HOMA-IR and HOMA-β. |
