| Functional gastrointestinal disorders(FGIDs)is a group of disease in gastroenterology,among which the most two common types are functional dyspepsia(FD)and irritable bowel syndrome(IBS).And the abnormal secretion of brain-gut peptide may be the mechanism.While as crucial brain-gut peptides,the expression level of corticotrophin-releasing factor(CRF)and anorexic peptide(Nesfatin-1)and their role in FGIDs remains largely unknow.Objective: To investigate the expression levels of CRF and Nesfatin-1in the serum samples of FD and IBS patients,explore the relationship between the expression levels of serum CRF and Nesfatin-1 and psychosocial factors and the severity of gastrointestinal symptoms in patients with FD and IBS,and analyze the correlation between CRF and Nesfatin-1.Methods: Thirty cases of FD patients(FD group),42 IBS cases of patients(IBS group)and 20 cases of healthy subjects(N group)were included.Self-rating anxiety scale(SAS)and self-rating depression scale(SDS)were used to assess the anxiety and depression status of patients.According to the results of SAS and SDS,the FD group was further divided into the anxiety/depression group(FD-A group)and the non-anxiety and depression group(FD-B group);the IBS group was divided into the anxiety/depression group(IBS-A Group)and the group without anxiety and depression(IBS-B group).The expression of CRF Nesfatin-1 in serum was determined by enzyme-linked immunosorbentassay.Gastrointestinal Symptom Rating Scale(GSRS)was used to assess the severity of gastrointestinal symptoms.Pearson correlation analysis was used to evaluate the correlation between serum CRF,Nesfatin-1 expression levels and SAS scores,SDS scores,and GSRS scores,and the correlation between serum CRF and Nesfatin-1 expression levels.Result:1.Expression level of serum CRFThe expression level of CRF in the FD group was significantly higher than that in the N group(45.089±11.091 VS.31.129±5.659,P<0.05).The expression level of CRF in the FD-A group was significantly higher than that in the FD-B group(50.049±10.305 VS.38.604±8.675,P<0.05).The expression level of CRF in the FD-B group was significantly higher than that in the N group(38.604±8.675 VS.31.129±5.659,P<0.05).The expression level of CRF in IBS group was significantly higher than that in N group(51.722±14.080 VS.31.129±5.659,P<0.05).The expression level of CRF in the IBS-A group was significantly higher than that in the IBS-B group(59.449±11.360 VS.40.359±9.163,P<0.05).The expression level of CRF in the IBS-B group was significantly higher than that in the N group(40.359±9.163 VS.31.129±5.659,P<0.05).2.Expression level of serum Nesfatin-1The expression level of Nesfatin-1 in the FD group was significantly higher than that in the control group(175.631±30.836 VS.136.650±12.631,P<0.05).The expression level of Nesfatin-1 in the FD-A group was significantly higher than that in the FD-B group(187.312±33.083 VS.160.356±19.812,P<0.05).The expression level of Nesfatin-1 in the FD-B group was significantly higher than that in the N group(160.356±19.812 VS.136.650±12.631,P<0.05).The expression level of Nesfatin-1 in the IBS group was significantly higher than that in the N group(191.970±46.946 VS.136.650±12.631,P<0.05).The expression level of Nesfatin-1 in IBS-A group wassignificantly higher than that in IBS-B group(205.33±24.465 VS.172.32±35.577,P<0.05).The expression level of Nesfatin-1 in the IBS-B group was significantly higher than that in the N group(172.320±49.647 VS.172.320±12.631,P<0.05).3.Correlation analysis of serum CRF expression level with SAS score,SDS score,and GSRS scoreThe serum CRF expression level in the FD group was positively correlated with the SAS score(r=0.710,P<0.01),positively correlated with the SDS score(r=0.681,P<0.01),and positively correlated with the GSRS score(r=0.687,P<0.01).The CRF expression level in the IBS group was positively correlated with the SAS score(r=0.858,P<0.01),positively correlated with the SDS score(r=0.830,P<0.01),and positively correlated with the GSRS score(r=0.748,P<0.01).4.Correlation analysis of serum Nesfatin-1 expression level with SAS score,SDS score,and GSRS scoreThe serum Nesfatin-1 expression level in the FD group was positively correlated with the SAS score(r=0.558,P<0.01),positively correlated with the SDS score(r=0.559,P<0.01),and positively correlated with the GSRS score(r= 0.566,P<0.01).The serum Nesfatin-1 expression level in the IBS group was positively correlated with the SAS score(r=0.654,P<0.01),positively correlated with the SDS score(r=0.678,P<0.01),and positively correlated with the GSRS score(r= 0.491,P<0.01).5.The expression of serum CRF in patients with FD and IBS was positively correlated with the expression level of Nesfatin-1(r=0.666,P<0.01;r= 0.730,P<0.01).Conclusions:CRF and Nesfatin-1 are highly expressed in serum samples of FD and IBS patients,and the increased expression level is related to the anxiety anddepression state and the severity of gastrointestinal symptoms of FD and IBS patients.CRF and Nesfatin-1 may involve in the development of FD and IBS. |
