ObjectiveChanges in serum amylase and lipase levels are commonly used to evaluate the prognosis of acute pancreatitis.The specificity and accuracy of these two clinical indicators are not high enough.Therefore,they can only be used as auxiliary diagnostic markers.Many biomarkers have been studied as potential predictors of the severity and prognosis of acute pancreatitis.However,due to low accuracy,tedious laboratory techniques and high costs,new biomarkers have not been adopted clinically.Lipids,as an important constituent material basis of biofilms,play an important role in the processes of material exchange,energy transfer,signal pathway conduction,and apoptosis.Changes in the overall metabolic profile of lipids have been found in the pathological state of various diseases.The occurrence of acute inflammation is bound to affect key enzymes or mechanisms related to intracellular lipid metabolism.Therefore,liquid chromatography coupled to mass spectrometry(LC-MS)was applied to the acute pancreas in this study.Obtain metabolic profile data of inflammation samples,and combine it with lipidomics analysis method to obtain differential lipid metabolites between acute pancreatitis and healthy people.Provide a valuable reference for the future screening of differential metabolites that can identify acute pancreatitis and healthy people,and find potential prognostic markers for acute pancreatitis.MethodsThis experiment uses a case-control group analysis method,with acute pancreatitis as the case group,and cholecystitis and healthy people as the control group.HPLC single-stage quadrupole time-of-flight tandem mass spectrometry was used to detect lipid metabolomics in acute pancreatitis,cholecystitis,and healthy individuals.The differences of lipid metabolomics between the two groups were analyzed by one-dimensional and multi-dimensional statistical methods.Screen differential metabolites based on molecular mass,metabolite database,and mass / mass spectrometry(MS / MS)information.The area under the receiver operating characteristic curve(AUC)of differential metabolites was used to evaluate their value in identifying acute pancreatitis and healthy individuals.ConclusionsA total of 9 differential lipid metabolites were screened from the metabolomes of patients with acute pancreatitis,cholecystitis,and healthy individuals.The 9 lipid metabolites can be divided into 3 types:phosphatidylethanolamine,phosphatidylglycerol,and lysostearyl inositol.Category,9 differential lipid metabolites are significantly lower in pancreatitis than cholecystitis and healthy people.Among them,PE(O-16:0/18:2)has a good correlation with the disease changes of acute pancreatitis,and may be a potential prognostic marker for acute pancreatitis.
