Objective:Nowadays,the plastic surgery industry is developing vigorously and fat graft surgery is widely used.However,the high absorption rate and low survival rate of transplanted fat still limit the application of autologous fat grafting.Reports said that fat stem cells can effectively improve the survival rate of fat graft,but the application of adipose stem cells(ADSCs)still face many restrictions.It is a hot research topic to find one substitute that function like stem cells.Then we tend to extracellular vesicles(EVs).Due to the content of EVs and difficulties in obtaining them,we finally studied the umbilical cord stem cells(UCMSCs)in plastic surgery.Methods:In this study,ADSCs were extracted from adipose tissue obtained from autologous fat graft using hydrodynamic equipment,and UCMSCs were cultured.We aimed to verify the osteogenic and osteogenic differentiation of stem cells.In the first part,we extracted the EVs derived from ADSCs by classic differential centrifugation,and identified the EVs by electron microscopy,NTA and western blot.In the second part,adipose tissue mixed with EVs derived from ADSCs and adipose tissue mixed with UCMSCs were injected subcutaneously in the symmetrical area of the back of nude mice.Results:The results proved that EVs derived from ADSCs could effectively improve the survival rate of transplanted fat,UCMSCs can also effectively improve the survival rate of transplanted fat.The EVs and UCMSCs groups had higher expression of CD34,VEGF,KI67 than the control group.Conclusion:EVs can effectively improve the survival rate of transplanted fat and are related to the expression of VEGF.UCMSCs have a significant pro-vascular effect in fat transplantation,and VEGF and CD34 are significantly increased.Based on the above experimental results,we hope to obtain a safer and more effective way to improve the survival rate of fat grafting in basic research as well as clinical trials. |
