ObjectiveRheumatoid arthritis belongs to an autoimmune disease,often with multiple organs and organs outside the joint,dry eye disease(DED)is the most common eye disease of rheumatoid arthritis,and often due to The discovery and effective treatment lead to blindness in rheumatoid arthritis dry eyes,which seriously affects the quality of life of patients.Therefore,early detection of ocular lesions in patients with rheumatoid arthritis is very important for the treatment and prognosis of patients.In this study,label-free quantitative proteomics technology was used to detect serum proteomics in patients with rheumatoid arthritis and dry eyes,to screen for differentially sensitive proteins,and to analyze their functions and biological processes involved.Early serological markers of rheumatoid arthritis combined with dry eyes and a further basis for exploring the pathogenesis of rheumatoid arthritis dry eyes.MethodsFrom June 2018 to June 2019,20 patients with rheumatoid arthritis diagnosed or diagnosed in the rheumatology and immunology at Panjin Gem Flower Hospital were selected as the control group,including 7 men and 13 women.Rheumatoid arthritis(course of 1 to 20 years,average 10 years)with20 patients with dry eye as the observation group,including 4 males and 16 females,using label-free quantitative proteomics technology Serum proteins were accurately identified and quantified,and differentially expressed proteins were screened out.The functions of these differential proteins and the biological processes involved in the pathogenesis of RA dry eye were analyzed bybioinformatics techniques.ResultsIn this project,Label-free quantitative proteomics technology was used to carry out the project research.A total of 345 proteins were identified in the two comparison groups of rheumatoid arthritis and rheumatoid arthritis combined with dry eyes,and 13 differentially expressed proteins were screened,including upregulated protein 9 Peroxidase-2,serum amyloid A-1,islet regenerative protein 3-α,complement factor H-related protein 3,immunoglobulin mu heavy chain,immunoglobulin heavy variant 2-70 D,immunoglobulin Protein lambda variable 7-43,lipoprotein a,immunoglobulin lambda variable 3-21,four down-regulated proteins,namely pregnancy area protein,glycated hemoglobin,immunoglobulin heavy change 1-2,immunoglobulin delta heavy chain.13 different proteins play important roles in cellular metabolism,biological regulation,and biological processes,and have molecular binding functions.They participate in oxidative stress,immune and inflammatory reactions,lipid metabolism and other processes in the development of rheumatoid arthritis dry eyes.Conclusions1、Compared with the rheumatoid arthritis group alone,there are many different proteins in the rheumatoid arthritis combined dry eye group.2、Label-free quantitative proteome The serum differential proteins screened by biological technology,namely islet regenerative protein 3-α,complement factor H-related protein,pregnancy area protein,and glycated hemoglobin may participate in the pathogenesis of rheumatoid arthritis dry eyes,but can it be used as a diagnosis of rheumatoid joints The early serological markers of Yangan eye will be verified by further research.3、Peroxidase-2,serum amyloid A-1,related immunoglobulins,and lipoprotein a can be used as serological markers for early diagnosis of rheumatoid arthritis dry eyes.The research on eye pathogenesis indicates the direction. |
