| Objects To screen differentially expressed lnc RNAs in the development of RSV-induced asthma and investigate the possible molecular mechanism underlying.Methods Among HDC-CONTROL group,HDC-RSV group and HDC-BEAS+RSV group,we screened differentially expressed genes and lnc RNAs of dendritic cells(DCs)by Affymetrix Human Transcriptome Array 2.0.Then,we analyzed these lnc RNAs through cluster analysis and functional analysis.We detected the expressions of six differentially expressed lnc RNAs by quantitative real-time PCR(q PCR).We selected lnc RNA n337374 as the research objective.The m RNA expression of lnc RNA n337374 corresponding gene(n337374)in DCs infected by lentiviral vectors which excessively expressed lnc RNA n337374 was also detected.In addition,airway hyper-responsiveness of lnc RNA n337374-overexpression RSV induced asthmatic mice was measured using whole body plethysmograph.After lnc RNA n337374-overexpression DCs were co-cultured with RSV-infected bronchial epithelial cells,we compared the expression of five cells surface molecule of DCs by q PCR and flow cytometry.Then signaling pathways were analyzed by western blot.Results 1.Differentially expressed genes and lnc RNAs were screened out.Through the GO and pathway analysis,we discoverd up-regulated and down-regulated lnc RNAs and found that the up-regulated pathways were mainly related to immunoregulation.2.We found the m RNA expression of n337374 was less in HDC-BEAS+RSV group than that in other group(p<0.01).3.We detected the m RNA expression of n337374 in DCs was significantly more compared with uninfected group and control group(p<0.01).4.The specific airway resistance in different concentrations of aerosolized methacholine was decreased obviously in the asthmatic model which excessively expressed lnc RNA-n337374.5.The expression of CD86 in DCs was decreased after DCs which over-expressed lnc RNA n337374 were co-culcured with RSV-infected bronchial epithelial cells.And MAPK signaling pathway is the signaling pathway.Conclusion 1.The lung function were improved in asthmatic model induced by RSV which excessively expressed lnc RNA n337374;2.Lnc RNA n337374 regulated the maturation and function of DCs by downregulating CD86 through ERK1/2 signal pathway. |
