Spodoptera frugiperda is an omnivorous insect of the order Lepidoptera,which originated in tropical and subtropical America,and mainly damages corn.In recent years,Spodoptera frugiperda has invaded many countries in Africa and Asia,including China,and has seriously threatened the food security of newly invaded countries and regions.Cannibalism or cannibalism is a common phenomenon in the insect world.The larger individual often feeds on the smaller one when food is scarce.There is currently no in-depth research on this behavior.DNA/RNA methylation belongs to a kind of regulation mechanism of epigenetics,which is widely present in eukaryotic organisms.More and more studies have found that many biological processes are related to methylation modification.This study assumes that the level of methylation modification in insects changes from normal state to starvation to cannibalism.This study took Spodoptera frugiperda as the research object.By setting up experimental groups,it was first verified that it has the characteristics of cannibalism when lacking food,and the subsequent anatomical extraction experiments and the use of LC-MS/MS system to detect the characteristics of this type of insect The m6 A content of RNA in the midgut tissue was significantly different between different groups.The overall m6 A level of the starvation group was lower than that of the normal control group,and the overall m6 A level of the cannibalism group was higher than that of the starvation group.It is preliminarily judged that Spodoptera frugiperda has undergone RNA m6 A methylation during cannibalism.Subsequently,in order to conduct an in-depth study on the distribution and function of m6 A modification in this process,this experiment uses Me RIP-Seq/m6A-Seq technology to study the m6 A modification regions of the whole genome,and conducts bioinformatics analysis on the sequencing results obtained,and Statistical identification of sequencing data.The results showed that the distribution of m6 A in the gene elements of the normal group and the cannibalism group was almost the same,showing a regular level,and similar to mammals,mainly distributed in the exon region and 3’UTR region,while the starvation group m6 A in the gene There is a huge difference in the distribution of the components.The distribution in the 5’UTR area accounts for close to 40%,while the other two groups are only distributed around 5% in this area.The subsequent analysis results showed that the distribution and gene expression level of m6 A modification in the normal group and the cannibalism group were very similar,belonging to the normal level,while the starvation group had a big difference.In this study,the three sets of samples were compared in pairs,and finally the overlapping part of the differential m6A-related genes and differentially expressed genes was found.This part of the gene was screened,and the gene LOC118279736 is suspected to be cytochrome P450 6B7,which may be related to detoxification and feeding Functionally related.This gene can be used as a target gene for follow-up experiments such as knockdown and overexpression.By observing whether the phenotype after abnormal expression of methylases is restored,we can further explore whether the cannibalism behavior can be artificially regulated,which is important for the development of new pests.The prevention method has forward-looking value.
