Stress is pervasive in this world.Stress conditions can perform to several psychological and physiological disorders.Disorder of homeostasis called homeostasis and stress it is determined by a stimulus described as a stressor.In general,three kinds of stressors psychological,metabolic,and physical in researches on stressors are mostly used in a mixture e.g.restraint stress it is an integration of mental and physical.A huge variety of physiological behavioral and neuroendocrine changes happened to adaptability with stress events and the recovery.Stress considered as a factor to impact the digestion process,growth performance,feed and water consumptions,various effects on gastrointestinal tract,reproduction adversely,and many other organs in the body.Resveratrol is a compound found in grapes and wine.It has been mentioned a several benefit for Resveratrol such as antioxidant,anticancer,anti-inflammatory,anti-aging,and cardioprotective effects.1.Protective roles of Resveratrol against restraint stress effects in adult stomach of male miceTo investigate if restraint stress has negative effects on the stomach of mice(Mus musculus)while applying Resveratrol treatment,adult male mice(25-30 g)divided into 5 groups as follow;control group(C),restraint stress group(RS),restraint stress with vehicle group(RS+V),RS with 2mg Resveratrol group(RS+2mg RES),and RS with 20mg Resveratrol group(RS+20mg RES).The mice were restrained in conical centrifuge tubes for four hours daily to establish RS model.The experimental groups RS+2mg RES,RS+20mg RES and RS+V were given oral dose of Resveratrol or vehicle for fifteen days.The results indicated no change was found in the water and feed consumptions in control group.In contrast,RS,RS+V,and RS+20mg RES groups were significantly reduced the consumptions of feed and water compared with control group,while,the group of RS+2mg RES was insignificantly in consumptions of feed and water.The control group mice showed increasing in the body weight during the experiment compared to RS and RS+V groups which showed a significant reduction.In contrast,groups treated by Resveratrol(RS+2mg RES and RS+20mg RES groups)showed significant improvements in the body weight gain compared to RS and RS+V groups.For further,RS and RS+V and RS+20mg RES groups got acute damage on the morphology of gastric cells and reduces the quantitative and distribution of parietal cells,along with the decreased size and diameter as compare to control group which was showing normal histology,and stressed group RS+2mg RES was in normal histology too.Furthermore,we observed that the antibody ofinducing factor(AIF)was presented and attached highly in RS and RS+V groups as compare to control group,while AIF antibody attached normally in stressed groups RS+20mg RES and RS+2mg RES as compare to control group which pointed to ulceration.Our finding has proved the significant alleviative effects of Resveratrol against restraint stress in the mouse stomach.2.Protective roles of Resveratrol against restraint stress effects in spleen of adult male miceThis study used fifty adult male mice(25-30 g)divided into 5 groups as follow;control group(C),restraint stress group(RS).restraint stress with vehicle group(RS+V),RS with 2mg Resveratrol group(RS+2mg RES),and RS with 20mg Resveratrol group(RS+20mg RES).The mice were restrained in conical centrifuge tubes for four hours daily to establish RS model and investigate restraint stress negative effects on the spleen of mice while applying Resveratrol treatment.The experimental groups RS+2mg RES and RS+20mg RES were given oral dose of Resveratrol or vehicle for fifteen consecutive days.The results of the spleen size,weight,and length in the control group showed no change during the experiment.While,RS,RS+V and RS+20mg RES groups significantly increased the size,weight,and length of spleen as compared to the control group.RS+2mg RES group insignificantly increased size,weight,and length.In addition,we have selected 3 groups as follow;control,RS,and RS+2mg RES from five experiment groups for the histological examination(H&E)stressed group RS got acute damages on the morphology of spleen structure and reduces the quantitative and distribution of spleen components.While these phenomen were not present in control and RS+2mg RES groups.Our finding has proved that the restraint stress can lead to splenomegaly and Resveratrol can improve the health of spleen and control the negative results.In conclusion,our research result showed that resveratrol can improve the general health in stressed mice,also has protective roles from negative effects of restraint stress in stomach and spleen of adult male mice.
