| Taenia solium is one of the major foodborne parasites,which is identified as one of the key zoonotic diseases to be studied and controlled.at the same time,it is also listed as "one of 17 neglected tropical diseases" by the World Health Organization.The different stages of Taenia solium development have complex effects on the host,which are mainly spread between the environment and the host through eggs or gestational nodes,and susceptible hosts and infected hosts are accompanied by inflammation.The eggs or gestational nodes are in different parts of the host’s body.Parasitic,adult parasites in the human intestines,other parts of the cysticercus stage exist,causing inflammation,cysticercus gradually grow up and form vesicles,so that the inflammatory response gradually disappears and escapes the immune mechanism.Cysticercus cellulosae has a long incubation period in the human body,and the parasitic part may cause disability and visual disturbance to a lesser extent,and cause ataxia,epilepsy and other brain diseases.In China,the prevalence of cysticercosis is mainly distributed in the northeast,southwest and remote mountainous areas of China.Unfavorable conditions such as geographical location,underdeveloped economy,and inconvenient transportation limit the diagnosis of cysticercosis with the help of proteomics technology.Cysticercus cellulosae and the common protein in the patient’s serum,in order to solve the current embarrassing situation of cysticercus cellulosae infection in remote areas.Label-free qualitative technology is a research method based on mass spectrometry proteomics,which is suitable for proteomics research of species with small sample size or rare samples.In order to find out the common protein between the cysticercosis patients and the cysticercosis patients,firstly use high performance liquid chromatography-tandem mass spectrometry system to perform qualitative analysis of the total protein of the worms and the patient’s serum,and then screen the source of pigs.The common protein of cysticercus body and patient serum.The results showed that,after human infection with Cysticercus spp.,the cysticerial protein stimulates the host’s immune response.Analyze porcine cysticercosis and patient serum proteomics through gene ontology(GO),orthologous cluster(COG),Kyoto Encyclopedia of Genes and Genomes(KEGG),etc.,and enrich to participate in intracellular processes and monomers Proteins such as process and biological regulation are mainly involved in signal pathways such as complement and coagulation cascade,pentose phosphate pathway,and ECMreceptor interaction.By predicting the domain of the worm body protein,the genes and Histonefold Domain of Histone H2A/H2B/H3,the conserved domain of the cysticercus structure,were found.High performance liquid chromatography-tandem mass spectrometry system identification and analysis showed that there is a common protein between swine cysticercosis and patient serum,namely A0A0R3X5M7 protein.In order to verify the accuracy of the identification results of Lable-free,the prokaryotic expression system was selected to induce the expression of the shared protein and further purified,the proteomics results were verified by the ELISA method,and the ELISA results were rechecked by the Western-blot technology.Western-blot results showed that the serum test results of Taenia swine,Trichinella swine,and Clonorchis sinensis were positive,and the coincidence rate with the ELISA results was 100%.The protein A0A0R3X5M7 screened by Lable-free proteomics research is a common protein of Cysticercus cellulosae and the patient,but it cannot be used as a diagnostic marker for the diagnosis of Cysticercus cellulosae.This experiment lays the foundation for other techniques of proteomics to study the effect of Taenia solium on the host body and to screen the diagnostic antigen of cysticercosis. |
