Radiation mutation breeding has been widely used due to its high mutation rate,high survival rate,wide mutation spectrum,and fast stability.However,most radiation mutation breeding studies still remain in the observation of apparent traits of mutagenic plants and the screening of beneficial mutants.Molecular biology studies that cause mutations are lagging behind.Light has a regulatory effect on plant growth,morphogenesis,metabolism and gene expression.The phytochrome senses light signals and continuously regulates plant physiology and development.There have been few reports on the coordination of radiation and light in mutagenic breeding.These have become two issues that are worthy of further study to cultivate new varieties of high quality plants.This work is based on kermes rice and Trollius Chinensis:heavy ion ~7Li radiation and different LED light conditions were used to study the effects of radiation and light on the relative expression of phytochrome genes and apparent traits in the kermes.The high-energy radiation and space-borne radiation methods were used to analyze the apparent traits and gene polymorphisms of the Trollius Chinensis,and to explore the effect of the different duration on its ionizing radiation effect.The radiation mutagenic effects of the two plants were studied in depth from the apparent traits of plants and the level of molecular biology,and the following innovative work was completed:1.The combination of radiation and light quality showed that the field characteristics and relative expression of phytochrome genes in kermes rice showed non-linear changes with the increase of radiation dose.Different light condition had different effects on the field characteristics and the relative expression of phytochrome gene in the M2 generation of kermes rice seedlings,and the red light promoted the elongation of the second internode of the M2 generation of kermes rice seedlings.2.Through space ionizing radiation analysis,it can be seen that there are certain doses and genetic polymorphism differences between the two batches of Trollius Chinensis with the same plant germplasm,the same flight orbit,and different flight time.3.~7Li heavy ion 10Gy radiation dose can significantly increase the production of kermes rice,fast neutron radiation can produce 10cm diameter of the Trollius Chinensis large flowers and other beneficial variations.Studies have shown that the selection of specific radiation doses is conducive to the cultivation of new varieties of high quality plants.
