Potato late blight is one of the most serious diseases in the world.It is called "potato cancer" because of its strong destructive power,which brings huge economic losses to agricultural production.We have been spraying chemical pesticide control potato late blight,which has caused serious environmental pollution and food security problems.Therefore,it is necessary to research and develop low toxicity and high efficiency biological pesticides.Ethylicin is the first new pesticide variety in China.It has the advantages of quick detection,low dosage and low cost.It has been widely used on fungal and bacterial diseases of various crops.It is of great significance to explore the inhibitory effect of ethylicin on potato late blight and analyze its antibacterial mechanism for the prevention and control of potato late blight.In this study,strains T30-4 and 88069 of Phytophthora infestans(P.infestans)were used as research subjects to analyze the inhibitory effect of ethylicin on the growth,sporulation,spore germination and virulence of P.infestans.Proteomics and metabonomics analysis were carried out to study the bacteriostasis mechanism after treatment with ethylicin.The following experimental results were obtained:(1)The results showed that ethylicin could inhibit the mycelial growth,sporulation ability,spore germination and pathogenicity of P.infestans T30-4 and 88069 strains,and had a dose-dependent effect,indicating that ethylicin had a certain inhibitory effect on potato late blight.(2)Using half effective inhibition concentration(IC50)concentrations of ethylicin treat T30-4 strains,metabolomics analysis showed that ethylicin treatment T30-4 strains caused by disease,the most significant change differentially expressed metabolites are amino acids,the most significantly changed metabolites amino acids were and expression levels were significantly down-regulated,indicating that ethylicin significantly inhibited amino acid metabolism of P.infestans.Due to amino acids serve as the substrates and basic component of proteins,ethylicin may inhibit protein synthesis in pathogens by inhibiting amino acid metabolism.(3)Proteomics analysis,GO and KEGG pathway analysis found that the ribosomal changes were the most significant,and the expression levels of differentially expressed proteins enriched in ribosomes were most down-regulated,indicating that ethylicin significantly inhibited the ribosomal function of pathogenic bacteria.The ribosome is the site of protein synthesis,so ethylicin may inhibit protein biosynthesis by inhibiting ribosome function in pathogens.In addition,BCA protein quantification further confirmed the results of proteomics analysis.(4)After treated by ethylicin,use equipment to measure various indicators including the potato plant’s height,stem diameter,number of nodules and number of leaves.The results showed that ethylicin had no effect on the normal growth of potato plants.The potted experiment showed that ethylicin had a certain inhibitory effect on potato late blight,and field experiment showed that the combination of ethylicin and infinito could reduce the use of chemical pesticides and achieve the same control effect.In conclusion,ethylicin can have a certain inhibitory effect on potato late blight,and according to proteomics and metabolomics analysis,ethylicin mainly inhibits protein synthesis of by inhibiting ribosome function and amino acid metabolism,and finally achieves the effect of inhibiting potato late blight.
