| F.oxysporum f.sp.benincasae is a sharp knife scythe wax gourd type specialization caused by F.oxysporum f.sp.benincasae Gertagh M&Ester A,has been a serious threat to Benincasa hispida(Thunb.)Cogn production,solve the winter melon wilt disease is the most simple and effective method is using the scion grafting cultivation of resistance,but the anvil with wax gourd varieties of resistance to blight,rarely reported in urgent need of appropriate winter melon grafting anvil with winter melon germplasm screening and resistance identification,etc.In this study,we selected the best inoculation concentration for the identification of Fusarium wilt resistance of winter gourd from 12 anvil germplasm resources preserved by our research group,and identified the disease resistance of these anvil winter gourd.Illumina Hi Seq2500 high-throughput sequencing platform was used to analyze the whole-genome expression profiles of resistant and susceptible varieties of winter gourd Fusarium wilt before and after inoculation,and some differential genes with high correlation with Fusarium wilt resistance of winter gourd were obtained.Further,12 samples of winter gourd germplasm used as stock and’black iron 999’were used as scion.To investigate the survival rate of grafted seedlings,the field growth and development of grafted plants,yield and fruit quality,etc.,laid a theoretical foundation for the molecular mechanism of resistance to Fusarium wilt of waxgourd for anvil use and the study of grafting suitability of waxgourd.The main results of this study are as follows:.1.Through the screening test of inoculation concentration,the optimal inoculation concentration was 1×106cfu·m L-1,and the optimal inoculation amount was 5m L.Adopts the optimized system 12 anvil with winter melon germplasm identification,two germplasm(D01 and D02)characterized by Gao Gan,four of the germplasm(D03 D04 D05 and D08)characterized by disease,three species(D06,D07 DC02)and three copies of performance for the resistance,three species(D09,DC01 and DC03)for disease resistance,including DC03 disease index,lowest 26.25.2.The survival rates of 11 germplasms(D01,D02,D03,D04,D05,D06,D07,D08,D09,DC02 and DC03)grafted with white gourd’Hei Tie 999’were all higher than 90%,which could be used as candidate parents for grafting stock.Grafting 11 germplasm with high survival rate,there are 8 species(D01,D02 D03 D04,D05,D06,DC02 DC03)and the grafting yield was significantly higher than that of CK,compared with the CK plant yield were increased by78.07%,42.16%,38.31%,10.06%,6.34%,13.84%,46.27%,19.21%and49.62%,and that they have the potential to increase,can be used as a root stock selection of candidate of production;Four germplasm(D01,D03,D05,D06 and D08)had better performance than self-rooted seedlings at seedling stage,and could be used as candidate parents for breeding new stock varieties with enhanced seedling characters.Six germplasm(D03,D04,D06,D08,D09 and DC03)grew better than self-rooted seedlings after transplanting,and could be used as candidate parents for the selection of new rootstock varieties to improve plant growth.The fruit nutritional quality of 8 germplasm(D02,D03,D05,D06,D07,D08,D09 and DC03)grafted seedlings was better than that of self-rooted seedlings,which could be used as candidate parents for the breeding of new stock varieties to improve fruit nutritional quality.Six germplasm(D01,D02,D03,D04,D06 and DC02)grafted seedlings had higher content of mineral elements in their fruits than self-rooted seedlings,which could be used as candidate parents for breeding new rootstock varieties to improve the mineral content of fruits.3.By transcriptome sequencing of resistant and susceptible wax gourd fusarium wilt germplasm before and after inoculation,a total of 167.24Gb of Clean Data were obtained,and the Clean Data of each sample reached 5.69Gb,and the percentage of Q30 base was 94.18%or above.Unigene was annotated and compared with KEGG database.By pattern clustering and enrichment analysis of differential genes,it was found that the major differentially expressed genes were concentrated in the metabolic pathways such as plant hormone signal transduction,plant-pathogen interaction,phenylpropanol synthesis and phenylalanine metabolism.Further through the q RT-PCR for styrene acrylic element on the biosynthesis of two differentially expressed genes(Bch09G003980、Bch03G023730),phenylalanine metabolic pathways of two differentially expressed genes(Bch03G012960、Bch08G003250),plant pathogen interactions on the path of two differentially expressed genes(Bch03G012960、Bch08G003250),plant hormone signal transduction of two differentially expressed gene(Bch01G013910、Bch10G015290)expression level quantitatively,The results showed that the trend of the expression of these genes by fluorescence quantification was consistent with that by transcriptome sequencing.Fusarium wilt resistance of anvil gourd germplasm may be the result of the combined action of phenylpropanol synthesis and phenylalanine metabolism. |
