The purpose of this experiment is to study the influence of the difference of intestinal microbes of purebred Kele pigs,Berkshire×Kele hybrid pigs and Suitai×Kele hybrid pigs,and the difference of intestinal microbes on the resistance to rough feeding.The content of SCFA in colon contents,the relative expression of SCFA receptors(GPR41 and GPR43)and transporters(MCT1 and MCT4)m RNA in colon tissues were measured,and the rough feeding resistance ability was compared and analyzed,which laid the foundation for the screening of hybrid combinations.The main findings are as follows:1.The richness of the jejunal flora of Suitai×Kele hybrid pigs is higher than that of pure-bred Kele pigs and Berkshire×Kele hybrid pigs,and the ileal flora of Suitai×Kele hybrid pigs is richer than pure-bred Kele pigs;Firmicutes and Proteobacteria is the dominant phyla in the small intestine of pure-bred Kele pigs,Berkshire×Kele hybrid pigs,and Suitai×Kele hybrid pigs.Lactobacillus is the dominant genus in the small intestine of the three.2.Firmicutes and Bacteroides are the dominant colonic phyla of pure-bred Kele pigs,Berkshire×Kele hybrid pigs and Suitai×Kele hybrid pigs.Prevotella,Streptococcus,Treponema,Alloprevotella and Rumenococcusare found in the colon of the three pigs.The relative abundance of Selenomonas in the colon of Berkshire×Kele hybrid pigs were significantly higher than those of pure-bred Kele pigs(P<0.05),and the relative abundances of Peptococcus,Phascolarctobacterium,Acetitomaculum and Leeia were significantly lower than those of pure-bred Kele pigs(P<0.05);the relative abundance trend of Lachnospira in the colon of pure-bred Kele pigs is higher than that of Berkshire×Kele hybrid pigs(P=0.056);Peptococcus and Phascolarctobacterium The relative abundance trend in the colon of Suitai×Kele hybrid pigs was higher than that in Berkshire×Kele hybrid pigs(P=0.06,P=0.09);the relative abundance trend of Peptococcus and Butyricicoccus in the colon of pure-bred Kele pigs was higher than that of Suitai×Kele hybrid pigs(P=0.0502,P=0.078).The colonic flora of purebred Kele pigs and Berkshire×Kele hybrid pigs was analyzed by LEf Se.The colon of purebred Kele pigs was enriched with a large number of short-chain fatty acid-producing bacteria.Among them,Acetitomaculum,uncultured_rumen_bacterium_4C0d_17,Leeia,Bacteroidia,Bacteroidales,belong to fiber-degrading bacteria,and some bacteria and synergistic effects with the fiber-degrading bacteria,they were Phascolarctobacterium and uncultured_Veillonellaceae_bacterium.The fiber-degrading bacteria such as Selenomonas and Clostridium leptum enriched in Berkshire×Kele hybrid pigs’ colon.The colonic flora of pure-bred Kele pigs and Suitai×Kele hybrid pigs was analyzed by LEf Se,Among them,Coprococcus,Bacteroides,Bacteroidia,Bacteroidales,Anaerostipes_hadrus,Sinorhizobium_sp_GL28,belong to fiber-degrading bacteria.The fiber-degrading bacteria Treponema_porcinum enriched in Suitai×Kele hybrid pigs’ colon.Comparing Berkshire×Kele hybrid pigs with Suitai×Kele hybrid pigs,the colony of Suitai×Kele hybrid pigs is enriched in the uncultured_Clostridium_sp of Terrisporobacter.It shows that the proportion of pure-bred Kele pig fiber degrading bacteria in colon is the highest,followed by Suitai×Kele hybrid pigs,and Berkshire×Kelehybrid pigs are lower.The content of short-chain fatty acids in the contents of the colonic intestine was determined,and there was no significant difference between the purebred Kele pigs,the Berkshire×Kele hybrid pigs and the Suitai×Kele hybrid pigs(P>0.05)3.The expression levels of GPR41 and GPR43 genes in the colon tissues of pure-bred Kele pigs were significantly higher than those of Berkshire×Kele hybrid pigs(P<0.05),and the expression levels of MCT1 gene in colon tissues of pure-bred Kele pigs were significantly higher than that of Berkshire×Kele hybrid pigs and Suitai×Kele hybrid pigs(P<0.05).In summary,the relative abundance of fiber-degrading bacteria in the colon of pure-bred Kele pigs,Berkshire×Kele hybrid pigs and Suitai×Kele hybrid pigs showed differences,with pure-bred Kele pigs being the highest,followed by Suitai×Kele hybrid pigs,and Berkshire×Kele hybrid pigs are lower.The expression of SCFA absorption and utilization related genes in the colon of pure-bred Kele pigs was significantly higher than that of Berkshire×Kele hybrid pigs and Suitai×Kele hybrid pigs,and there was no significant difference in gene expression between Berkshire×Kele hybrid pigs and Suitai×Kele hybrid pigs.It shows that the purebred Kele pigs have the strongest resistance to rough feeding,and the Suitai×Kele hybrid pigs have a stronger ability to digest crude fiber than the Suitai×Kele hybrid pigs.
