Flax is a unique oil crop in China,and its seeds are rich in unsaturated fatty acids.In this study,using transcriptome sequencing technique,the capsules of three flax varieties with different linolenic acid content(FR46,NY,FR48)were sampled and sequenced in three periods(0d,15 d and 30d).Through functional annotation and differential gene enrichment analysis,FPKM expression calculation and real-time fluorescence quantitative PCR verification were used to infer the candidate genes involved in linolenic acid synthesis and metabolism in flax.At the same time,the regulatory gene GDSL lipase family genes were identified,and transcriptome data were used to screen gene data sets to explore the regulatory role of GDSL lipase family genes in flax.In order to reveal the transcriptional regulation mechanism of flax fatty acid synthesis pathway,provide candidate genes for the genetic improvement of flax fatty acids,enrich the research theory of flax genomics.The results are as follows:1.15 samples of flax capsule formation were sequenced by transcriptome,and the sequencing quality was highly related to the reference genome and reliable.Transcriptome sequencing data showed that the change of DEGs in the three developmental stages of flax was greater than that among varieties.There were significant differences in the relative expression levels of most genes in the middle and later stage of capsule development.2.Based on the functional annotation of genes,the enrichment analysis of differential genes was carried out,the enrichment categories and expression profiles of differential genes were analyzed,and the core genes involved in fatty acid biosynthesis pathway in flax were explored.Nineteen candidate genes involved in fatty acid biosynthesis pathway were screened,and the expression of 15 genes were verified by real-time fluorescence quantitative PCR.Most candidate genes have higher expression levels in the later stage of capsule development.3.20 GDSL lipase genes in flax were identified by GDSL conserved sequence,and the identified family genes were analyzed by structure analysis and conservative motif analysis.Based on the previous transcriptome sequencing data and real-time fluorescence quantitative PCR verification,the expression level of GDSL lipase in different varieties and different developmental stages was analyzed,and the functional pathway of GDSL lipase was analyzed.It was found that Lus_GLEAN_10025368 and Lus_GLEAN_10043961 genes would regulate the oil metabolism of flax.
